Objective:Ovarian cancer is one of the three most common malignant tumors in women’s repro-ductive system,and the mortality rate ranks first among gynecological malignant tumors.Because of the concealed early lesions and the unclear clinical symptoms,most patients have entered the advanced stage of treatment,and the late cases lack effective treatment methods.At present,the 5-year survival rate of ovarian cancer patients is about 45%.Ther-efore,ovarian cancer is a serious threat to the life and health of Chinese women.At prese-nt,the etiology and mechanism of ovarian cancer are not very clear.Studies have shown that abnormal apoptosis of ovarian cancer cells is one of the important factors leading to the occurrence and development of ovarian cancer.ETS homologous factor(EHF),also known as epithelial specific ETS-3(ESE-3),belongs to the E26 transformation specific(ETS)transcription factor family.EHF transcription factors selectively bind to conserved ETS binding sites in target gene promoters to activate or inhibit gene expression involved in various biological processes,including cell cycle control,cell migration,cell proliferati-on,differentiation,apoptosis and angiogenesis.Studies have confirmed that EHF is involv-ed in the occurrence of various malignant tumors and plays an important role in regulating cell cycle and cell apoptosis.Wild-type p53-induced phosphatase-1(Wip1)gene is a p53-dependent proto-oncogene,which is widely involved in regulating a variety of cell signali-ng pathways and plays an important role in the occurrence of ovarian cancer and other can-cers.Wip1 is a serine/threonine protein phosphatase encoded by PPM1D gene,located in the 17q23/q24 region of human chromosome,and is a member of the PP2C family.Wip1gene is closely related to apoptosis.Some studies have found that Wip1 is involved in the process of inhibiting apoptosis of cancer cells,and the elevated expression of Wip1 gene has the effect of inhibiting apoptosis.At present,there are few reports about the expression and mechanism of EHF and Wip1 in epithelial ovarian cancer at home and abroad.In this paper,the expression of EHF and Wip1 in epithelial ovarian cancer was detected by immunohistochemical staining and Western blot.To further explore the relationship between EHF and Wip1 expression and epithelial ovarian cancer,and the correlation between EHF expression and Wip1 expressi-on in epithelial ovarian cancer,in order to clarify the significance of EHF and Wip1 in the occurrence and development of epithelial ovarian cancer.Method:105 paraffin-embedded ovarian tissues(65 cases of epithelial ovarian cancer,20 cases of benign ovarian tumors,20 cases of normal ovarian tissues)and 50 cases of fresh ovarian tissues(20 cases of epithelial ovarian cancer,20 cases of benign ovarian tumors,10 cases of normal ovarian tissues)were selected and divided into three groups.Immunohistochem-istry and Western bolt were used to detect the expression of EHF and Wip1 in normal ovar-ian tissue,benign ovarian tumor tissue and epithelial ovarian cancer tissue,and to analyze the relationship between the positive expression rate of these two factors and the clinicopa-thological characteristics of patients with epithelial ovarian cancer,as well as the correlati-on between the expression of EHF and Wip1 in epithelial ovarian cancer.Data were analyzed by SPSS20.0 software and Image J software.Chisquare test,Spe-arman correlation analysis and other statistical methods were used for statistical analysis.P<0.05 showed that the difference was statistically significant.Result:1.The expression of EHF was positioned in the nucleus.The positive rates of EHF in epithelial ovarian cancer,benign ovarian tumors and normal ovarian tissues were 76.92%,50%and 20%respectively.Its expression in epithelial ovarian cancer was significantly hi-gher than that in benign ovarian tumors and normal ovarian tissues(x~2=22.067,P=0.000).Further analysis showed that the expression of EHF in epithelial ovarian cancer was not re-lated to age,histological type,ascites and lymph node metastasis,but to clinical stage and histological grade.2.The expression of Wip1 was positioned in the nucleus and cytoplasm.The positive rates of Wip1 in epithelial ovarian cancer,benign ovarian tumors and normal ovarian tissu-es were 70.8%,40%and 15%,respectively.Its expression in epithelial ovarian cancer was significantly higher than that in the other two groups(x~2=25.736,P=0.000).Meanwhile,th-e expression of Wip1 in epithelial ovarian cancer was not related to age,histological type and ascites,but closely related to clinical stage,histological grade and lymph node metast-asis.3.The expression level of EHF protein in epithelial ovarian cancer(0.82±0.04)was significantly higher than that in benign ovarian tumors(0.49±0.06)and normal ovarian tissues(0.17±0.08),and the difference was statistically significant(P<0.05).The expressi-on level of Wip1 protein in epithelial ovarian cancer(0.79±0.04)was also higher than that in benign ovarian tumors(0.36±0.09)and normal ovarian tissues(0.18±0.07).The differe-nce was statistically significant(P<0.05).4.Spearman grade correlation analysis:EHF was positively correlated with Wip1 ex-pression(r_s=0.565,P=0.000).Conclusion:1.The expression of EHF and Wip1 in epithelial ovarian cancer was significantly hig-her than that in normal ovarian tissues and benign ovarian tumors.The high expression of EHF and Wip1 in epithelial ovarian cancer was closely related to the clinical stage and his-tological grade of patients.It is speculated that EHF and Wip1 may be involved in the occ-urrence and development of epithelial ovarian cancer.2.There was a positive correlation between EHF and Wip1,suggesting that the expre-ssion of EHF and Wip1 was synergistic.3.EHF and Wip1 are expected to become new targets for early detection and treatme-nt of epithelial ovarian cancer,and provide a new direction for diagnosis,treatment and pr-ognosis evaluation of epithelial ovarian cancer.
