| ObjectiveTo investigate the biological characteristics of infrapatellar fat padderived mesenchymal stem cells in synovial fluid from osteoarthritis patients.MethodsIPFP-MSCs were isolated and cultured in vitro,and stimulated by DMEM/F-12 medium,growth medium,or OA synovial fluid conditioned medium.The proliferation of IPFP-MSCs was detected by CCK-8 assay and the crystal violet staining assay.The senescence of IPFP-MSCs were detected by SA-β-gal staining.The migration and invasion of cell were detected by cell scratch test and Transwell assay.The expression of PCNA and CXCR4 was observed by indirect immunofluorescence,Western blot and real-time quantitative PCR.IPFP-MSCs were stimulated by chondrogenic induction medium with DMEM/F-12 medium,chondrogenic induction medium or chondrogenic induction medium with OA synovial fluid and compared for cell differentiation potential by using alcian blue staining.ResultsCompared with treated by DMEM/F-12 medium,the conditioned medium promoted the proliferation and migration of IPFP-MSCs,reduced the rate of SA-β-gal positive cell.The gene expression of PCNA increased and CXCR4 decreased in the group of conditioned medium.The supplementation of the induction medium with synovial fluid from OA induced IPFP-MSCs to differentiate into chondrogenic lineage.ConclusionOA synovial fluid can promote the proliferation and migration of IPFP-MSCs,it also play a positive assistance role in chondrogenic differentiation of IPFP-MSCs. |
