Objective:This study uses high-performance liquid chromatography-mass spectrometry(HPLC-Q-TOF-MS)to explore the metabolic differences between the serum of hepatocellular carcinoma and the serum of healthy subjects,and to find potential biomarkers to assist in the diagnosis of hepatocellular carcinoma,Explore the pathogenesis and treatment strategies of hepatocellular carcinoma.Method:HCC serum samples from patients admitted to the Department of Hepatobiliary Surgery of Hunan Provincial People’s Hospital from October 2019 to February 2020 were collected.All samples were 40 cases of HCC serum samples confirmed by pathology,and 40 cases of healthy subjects were collected.HPLC-Q-TOF-MS technology platform was used to perform multi-dimensional statistical analysis of differential metabolites in HCC serum and serum samples from healthy subjects.Metaboscape 3.0 software was used to preprocess the original metabolomics data,including peak extraction and normalization.Sim CA-P14.1 software was used for statistical analysis,and the differential metabolites of HCC cancer group and healthy subjects were screened according to the criteria of variable weight value(VIP >1),T test(P < 0.05)and FC > 1.5.Metabolite pathway analysis was performed using Metaboanalysis 5.0 to look for metabolic pathways associated with HCC.Result:1.The metabolic characteristics of HCC were obtained based on HPLC-Q-TOF-MS.Significant differences in metabolic fingerprints between HCC serum and healthy subjects were found,and a recognition model for distinguishing HCC from healthy subjects was successfully constructed.2.The study found significant differences in serum metabolic profiles between the HCC group and the Control group.Combined with VIP >1,FC >1.5 and P < 0.05,83 potentially different metabolites were screened out.Compared with the Control group,the serum levels of 37 metabolites in the HCC group were significantly increased,including vitamin D23 3-glucuronic acid,acetoacetic acid,L-glutamic acid,glycine,Lyso PC(24:0/0:0),cholesterol fatty acid ester(17:1).However,46 metabolites in the HCC group,such as retinoyl β-glucuronic acid,LYSOPC(18:1(11Z)/ 0:0),LYSOPC(16:0/0:0),glycerol phosphocholine,L-histidine,alanine and lactose,showed a significant downward trend.3.The synthesis and degradation of ketone body,D-glutamic acid and glutamine metabolism,glyoxylic acid and dicarboxylic acid metabolism,histidine metabolism,alanine,glutamic acid and aspartic acid metabolism,the pentose phosphate pathway,butyrate,GSH metabolism,glycine,serine and threonine metabolism,arginine biosynthesis and metabolism of galactose,a total of 11 metabolic pathways may participate in the pathological process of HCC.Conclusion:This study initially explored the changes of metabolites in the pathological process of HCC.The progress of HCC mainly leads to abnormal metabolic pathways such as fatty acid metabolism,amino acid,sugar metabolism,and phospholipid metabolism.Amino acid metabolism,choline and phospholipid metabolism are involved in the pathological process of HCC.The research results lay the foundation for the later study of the pathogenesis of HCC.
