Objective:Rosa laevigata Michx,is a traditional Chinese medicine,which belongs to Rosaceae,recorded in Chinese Pharmacopoeia in 2020,and has long been used as a medicinal and edible resource in the folk.Rosa laevigata Michx has become an important variety of traditional Chinese medicine for scientific researchers because of its diverse chemical constituents and extensive pharmacological activities.In this thesis,the chemical constituents from Rosa laevigata Michx were studied,and the triterpenoids with novel structure were found.Methods:The 70%ethanol extract of Rosa laevigata Michx was obtained by reflux extraction and the extract was separated and purified by macroporous resin,silica gel column,Sephadex LH-20chromatography,ODS reversed-phase column chromatography and preparative high performance liquid chromatography to get the monomer compounds.The structures were elucidated by 1D/2D NMR,UV,CD and high resolution mass spectrometry.CCK8 experiment was used to screen the obtained compounds for antitumor activity in vitro,and the effects of monomer compounds on the viability of human liver cancer SMMC-7721 cells and Hep G2 cells,human gastric cancer SGC-7901 and human breast cancer MDA-MB-231 were evaluated.Results:In this paper,14 compounds were isolated and identified,including 9 ursutane triterpenoids,1 oleanane triterpenoid,2secotriterpenes and 2 flavonoids.Compounds 1*-8*are new compounds.Compound 11 and 14 was isolated from Rosa laevigata Michx for the first time.Compound 11 shows obvious cytotoxicity to human gastric cancer cell SGC-7901 and human breast cancer cell MDA-MB-231,the IC50value of compound 11 was less than 35μM,compound 3*and compound 12 shows certain inhibitory effect on the proliferation of human liver cancer cell Hep G2,the IC50values are 19.20 and 56.82.Compound 6*shows cytotoxic effects on human breast cancer cell MDA-MB-231.Conclusions:A total of 14 compounds were isolated and identified including 8 new compounds which belong to triterpenoids.Four compounds with inhibitory effect on the proliferation of some cancer cells were screened out.
