| Objective: To explore the acute toxicity of water extract of Fang Qidan Yi Fei Tongqiao prescription to mice according to the guidelines of general pharmacology research of traditional Chinese medicine and natural drugs,and to explore the effect of water extract of AR Yi Fei Tongqiao prescription on serum interleukin-4 and interferon-γ in AR rats.Methods:1.acute toxicity test:⑴ Selecting 30 KM mice of SPF grade,Half male and half female,Randomly divided into single administration group,double administration group and three administration group,Fasting 12 h,before administration One,two,three times,6 h,interval between two doses No food is available during the interval.Normal feeding after administration,7 d,of mice were observed continuously To record the performance and death of mice,Determination of half lethal dose of water extract from Qidan Yi Fei Tongqiao prescription(LD 50).⑵Maximum tolerance test,20 KM mice of grade SPF,half male and half female,were randomly divided into blank group and experimental group,fasting water before gavage 12 h,the experimental group was given Qidan Yifei Tongqiao water extract concentration 2.8 g/m L and administration volume 40 ml/kg,blank group to give the same volume of distilled water.The expression and death of 14 d mice were observed continuously after administration,and the changes of body weight were recorded 2 w after dissection.2.pharmacodynamics experiment :32 rats of grade SPF were randomly divided into blank control group,AR model group,traditional Chinese medicine group(Qidanfei Tongqiao water extract)group and western medicine(loratadine)group,Eight in each group,Half male and half female.Except for the blank control group,All the other three groups were sensitized by intraperitoneal injection of ovalbumin(OVA)and induced by bilateral nasal drip.After making the model,The traditional Chinese medicine group was given Qidan Yifei Tongqiao prescription water extract solution to the stomach,The western medicine group was given loratadine suspension,The model group was given the same amount of normal saline.After 14 d of continuous administration,Death to rats,To calculate rat organ index,The contents of interleukin-4 and γ-interferon in serum were detected by ELISA method.Results:1.acute toxicity test⑴The mice survived and the lethal dose was not measured LD 50,So 2.8g/ml was used as the maximum concentration to determine the maximum dose and the maximum tolerance.(2)Maximum tolerance testThere were no abnormal changes in spontaneous movement,good mental state,normal drinking water and feeding.A total of 2 animals died in the experimental group,and one mouse died immediately during gastric lavage.The main cause of death was incomplete fasting and severe gastric dilatation.The contributing cause was improper gastric lavage,and some of the solution entered the trachea,causing choking and choking.Weight: With the time of administration,the weight of mice in the experimental group and control group increased to different degrees,the weight of mice in the 2 groups was significantly different before administration,1 week after administration and 2 weeks after administration(P<0.05).Compared with the control group,there was no significant difference between the experimental group and the blank group(PP<0.05),suggesting that the water extract of Qifitong prescription had no toxic effect on mice within 2 weeks after administration.After dissection,there were no obvious hyperemia,bleeding and edema in the thymus,spleen,liver and kidney organs of mice.There was no significant difference between the experimental group and the control group(P>0.05).It was suggested that the water extract of Qidanyifei Tongqiao decoction had no obvious toxic effect on the thymus,heart,lung,spleen and kidney of mice.2.Pharmacodynamics Experiment⑴WeightAfter 2 weeks of treatment,the weight of rats in Chinese medicine group and western medicine group gradually increased.After 2 weeks of treatment,the weight of rats in Chinese medicine group and western medicine group gradually increased.There was no significant difference in body weight between the AR group,the Chinese medicine group and the western medicine group(P﹥0.05),indicating that ovalbumin intraperitoneal injection plus nasal stimulation had an effect on the body weight of rats.After 2 weeks of treatment,there was no significant difference in body weight between the Chinese medicine group and the western medicine group(P﹥0.05),suggesting that the two intervention methods had an effect on the body weight of AR rats.Two weeks after treatment,the weight of rats in Chinese medicine group and western medicine group was significantly different from that in AR model group(P<0.05),indicating that the two intervention methods could improve the weight of AR rats.There was no significant difference in body weight between western medicine group and traditional Chinese medicine group(P ﹥ 0.05),indicating that the effect of the two intervention methods on the body weight of AR rats was similar.(2)Nasal symptomatology scoreThere were significant differences between AR model group,Chinese medicine group,western medicine group and blank group before treatment(P<0.05),indicating that the model of allergic rhinitis was established successfully.After 2 weeks of treatment,there was no significant difference between Chinese medicine group,western medicine group and blank group(P ﹥ 0.05),suggesting that the two intervention methods could improve sneezing,runny nose and itching symptoms of AR rats.There was no significant difference between Chinese medicine group,western medicine group and AR model group before treatment(P>0.05).After 2 weeks of treatment,the score of Chinese medicine group and western medicine group decreased,which was significantly different from that of AR model group(P<0.05),suggesting that the two intervention methods had significant effect on sneezing,nose and nasal itching.There was no significant difference between western medicine group and traditional Chinese medicine group after 2 weeks of treatment(P>0.05),indicating that Qidan Yifei Tongqiao prescription water extract and loratadine had similar efficacy in improving symptoms of AR rats.⑶Cytokine(IL-4、IFN-γ)detectionThe level of serum IL-4 in the AR model group and the level of IFN-γdecreased significantly(all P<0.05)compared with the blank control group.The serum IL-4 and IFN-γ levels of rats in traditional Chinese medicine group and western medicine group were significantly decreased(all P<0.05),indicating that both Qidan Yifei Tongqiao decoction and loratadine could reduce serum IL-4 and increase IFN-γ content.There was no significant difference in serum IL-4、IFN-γ level between Chinese medicine group and western medicine group(P>0.05),indicating that the effect of two intervention methods on regulating serum IL-4、IFN-γ level of AR rats was similar.Conclusion1.according to the guidelines of general pharmacological research of traditional Chinese medicine and natural drugs,the maximum tolerance multiple of water extract of Qidan Yi Fei Tongqiao prescription to mice was 333.8 times,and no obvious acute toxicity was found in mice after gavage,suggesting that the drug was safe.2.water extract of Qidan Yifei Tongqiao prescription may play a role in the treatment of allergic rhinitis by reducing serum IL-4 content and up-regulating serum IFN-γ content.The effect of water extract and loratadine 3.Qidan Yifei Tongqiao prescription on regulating serum IL-4、IFN-γ content in allergic rhinitis rats was similar. |
