| Objective:By comparing the differences of Th17 and Treg cells’ specific transcription factors RORγt,Foxp3 gene and protein levels in the the experimental group before and after treatment,the healthy group and the healed group,we preliminarily explored some of the target and mechanism of "Fu Xie" based on "Liang Xue Xiaofeng Powder" in the treatment of recurrent psoriasis vulgare(blood heat syndrome).To preliminarily explore the partial immunological factors of the disease’s susceptibility to recurrence;The partial immunological material basis of the blood heat syndrome of this disease was preliminarily explored to provide quantifiable indexes for the TCM syndrome differentiation and classification.Methods:Ten patients who met the diagnostic of recurrent psoriasis vulgaris and were blood heat syndrome regard as experimental group,treated with Liangxuexiaofeng powder and petroleum jelly for 8 weeks.Psoriasis vulgaris reached the clinical cure standard for more than one year in 10 cases were cured group,10 healthy cases were healthy group;Fasting venous blood was drawn before and after treatment;Real-time PCR and WB were used to determine the levels of RORγt and Foxp3 in every groups,and the differences in groups were compared and analyzed.Result:1、After treatment,the levels of RORγt,Foxp3 in the experimental group were significantly changed(P<0.05,P<0.05,P<0.05,P<0.01);The PASI score in the experimental group was decreased(P<0.01).2、 RORγt:(1)Before treatment,there were no significant differences in gene and protein expression in the experimental group compared with the cured group(P>0.05,P>0.05),but there were significant differences compared with the healthy group(P<0.05,P<0.05).(2)The difference between the cured group and the healthy group was significant(P<0.05,P<0.05).(3)After treatment,the gene protein expression of the experimental group was not significantly different from cured group(P>0.05,P>05),but was significantly different from healthy group(P<0.05,P<0.05).3、 Foxp3:(1)Before treatment,there were no significant differences in gene and protein expression in the experimental group compared with the cured group(P>0.05,4、P>0.05),but there were significant differences compared with the healthy group(P<0.05,P<0.05).(2)The difference between the cured group and the healthy group was statistically significant(P<0.05,P<0.05).(3)After treatment,the gene protein expression of the experimental group was not significantly different from cured group(P>0.05,P>05),but was significantly different from healthy group(P<0.05,P<0.05).5、Correlation analysis: There was no significant correlation between the levels of RORγt,Foxp3 gene and protein and PASI,pruritus and DQOLS in experimental group before and after treatment(P>0.05).Conclusion:Modified Liangxuexiaofeng powder plays a therapeutic role by enhancing the expression of RORγt gene and protein level and decreasing the expression of Foxp3 gene and protein level.After treatment,the PASI score was decreased.The high expression state of RORγt and low expression state of Foxp3 can be used as the microscopic material basis of the blood-heat syndrome of this disease,and may also be one of the immune factors for the susceptibility to recurrence of this disease. |
