Object:To explore the acupoint compatibility effect and the regulation mechanism,in HPA axis and SCN-PG-MT system,of HT7 and SP6 in improving daytime fatigue and sleepiness of insomnia.Methods:In this study,90 patients with insomnia were randomly divided into group of HT7,group of SP6 and group of HT7 and SP6,with 30 cases in each group.Electroacupuncture at the acupoint of HT7 in group of HT7,electroacupuncture at the acupoint of SP6 in group of SP6,and electroacupuncture at the acupoints of HT7 and SP6 in group of HT7 and SP6.The frequency of electroacupuncture was 5/25 Hz with a electric wave of low and high frequency.Treatment was performed once a day,and each treatment lasted for 30 minutes.The treatment lasted for 5 consecutive days as a course of treatment.Electroacupuncture therapy was performed for 2 courses in total,with 2 days’ rest between the courses.The Pittsburg Sleep Quality Index and Epworth Sleepiness Scale were used to evaluate the patients in the three groups on the next day of enrollment and the day after the treatment.Fasting venous blood was collected from the patients in the three groups,on the next day of enrollment and the day after the treatment,to detecte the contents of serum ACTH,CS and MT by enzym-linked immunosorbent assay(ELISA).Results:1.In this study,90 qualified cases were included,30 cases in each group,and 3 cases fell off,among which 1 case in each group were supplemented.There was no statistical significance in the general information and indicators of the three groups before treatment(P > 0.05).2.The total scores of PSQI of the group of HT7,group of SP6 and group of HT7 and SP6 after treatment was lower than that before treatment,with statistical significance(P<0.05);The total scores of PSQI of the group of HT7 and SP6 and the group of SP6 after treatment was lower than that of the group of HT7,with statistical significance(P<0.05);After treatment,scores of sleep quality,sleep time,sleep efficiency,sleep disorders and daytime dysfunction of the group of HT7 were lower than those before treatment,with statistical significance(P<0.05);After treatment,the scores of sleep quality,time before sleep,sleep time,sleep efficiency,sleep disorders and daytime dysfunction of the group of SP6 and the group of HT7 and SP6 were lower than those before treatment,with statistical significance(P<0.05);After treatment,the scores of sleep quality,sleep time and sleep efficiency of the group of HT7 and SP6 were lower than those of the group of HT7,and the scores of daytime dysfunction were lower than those of the group of HT7 and group of SP6,with statistical significance(P<0.05);After treatment,the scores of sleep quality and sleep efficiency of the group of SP6 were lower than those of the group of HT7,with statistical significance(P≤0.05).3.The total scores of ESS of the group of SP6 and the group of HT7 and SP6 after treatment was lower than that before treatment,with statistical significance(P<0.05).After treatment,the total scores of ESS of the group of HT7 and SP6 was lower than that of the group of HT7,with statistical significance(P<0.05).4.After treatment,the contents of serum ACTH and CS of the group of HT7,group of SP6 and group of HT7 and SP6 were all decreased compared with before treatment,with statistical significance(P<0.05);After treatment,the content of serum MT of the group of SP6 and the group of HT7 and SP6 was higher than that before treatment,with statistical significance(P<0.05);After treatment,the contents of serum ACTH and CS of the group of HT7 and SP6 were lower than those of the group of HT7,and the content of serum MT was higher than that of the group of HT7,with statistical significance(P<0.05);After treatment,the content of serum MT of group of SP6 was higher than that of group of HT7,with statistical significance(P<0.05).Conclusions:1.The compatibility of HT7 and SP6 has a synergism effect on the improvement of night sleep quality and daytime fatigue and sleepness of insomnia patients.SP6 has a better effect on the improvement of night sleep disorder and daytime fatigue and sleepness of insomnia patients than HT7,and plays a major role in acupoint compatibility.2.HT7-SP6 can downregulate the contents of serum ACTH and CS in patients with insomnia to inhibit the hyperactive HPA axis,increase the content of serum MT to improve the function of SCN-PG-MT system,and play the acupoint compatibility effect of improving sleep and daytime fatigue and sleepness.
