Objective:This thesis systematically studied the chemical constituents and biological activity of the dry stems and leaves from Uncaria rhynchophylla(Miq.)Miq.e x Havil.Methods:First of all,the high resolution liquid-mass spectrometry was used to analyze the material components of different parts of U.rhynchophylla and comparison of differences between medicinal and non-medical parts.Then,the chemical constituents of the non-medicinal part of U.rhynchophylla were isolated using silical gel,MCI,sephadex LH-20,Semi-prep HPLC and other separation methods respectively,and their structures were identified by MS,NMR and other spectral analysis methods.The MTT method was used to evaluate the neuroprotective activity of some monomer compounds.Results:The chemical compositions of chloroform extracts and ethyl acetate extract s in the drug and non-drug parts of U.rhynchophylla were analyzed,and their similarity and difference in composition type and distribution were obtained.A total of 26 compounds were isolated from the non-medicinal parts of U.rhy nchophylla.Structure elucidation was conducted via 1D NMR spectroscopic dat a interpretation,including 17 alkaloids,2 flavonoids,2 terpenoids,1 steroid,an d 4 other compounds.The compounds’name were hirsutine(LY-1),sucrose(LY-2),uncarialin(LY-3),dihydrositsirikine(LY-4),hirsuteine(LY-5),geissosch izine methyl ether(LY-6),indol[2,3-a]quinolizidine(LY-7),villocarine A(LY-8),dibutyl phthalate(LY-9),isorhynchophylline(LY-10),croceaine B(LY-11),4β-hydroxyisodolichantoside(LY-12),strictosamide(LY-13),cadambinic acid(LY-14),(-)epicatechin(LY-15),4-geissoschizine N-oxide methylether(LY-16),ursolic acid(LY-17),corynoxeine(LY-18),rhynchophylline(LY-19),β-amyrin-3-palmitate(LY-20),quercetin(LY-21),O-β-D-fructofuranosyl-(2→6)-α-D-gluco pyranosyl-(1→6)-β-D-fructofuranosyl-(2→6)-β-D-fructofuranosyl-(2→1)-α-D-gluco pyranosyl-(6→2)-β-D-fructofuranoside(LY-22),4s-corynoxeine N-oxide(LY-23),syringic acid(LY-24),vincosaminde(LY-25),p-hydroxybenzoic acid(LY-26).Compounds LY-2,LY-4,LY-7,LY-11,LY-12,LY-15,LY-16,LY-20,LY-22,LY-26 were isolated from U.rhynchophylla for the first time.Compounds LY-1,LY-3,LY-4,LY-5,LY-6,LY-7,LY-8,LY-16,LY-18,LY-19,LY-23,LY-25 were selected using MTT method to determine their effect on H2O2in duced nerve injury.The results show that 5 compounds(LY-16,LY-18,LY-19,LY-23,LY-25)have potential protective effects on H2O2-induced oxidative damage in SH-SY5Y cells.Conclusions:The above research results enrich the content of chemical constituents and biological activities of the non-traditional medicinal parts of U.rhynchophylla.The non-traditional medicinal parts and medicinal parts of U.rhynchophylla ha ve similarities in the types and distributions of chemical constituents.The use of non-traditional medicinal parts not only improve the utilization rate of U.rh ynchophylla,reduces resource waste,avoid environmental pollution,but also pro vide a scientific basis for the full utilization of Guizhou Uncaria resources.
