| Objective:Citri reticulatae pericarpium is one of the classic aging Traditional Chinese Medicine.However,according to the Chinese Pharmacopoeia,it has no meaning of aging.Most of the orange peel used clinically has certain dry side effects.After steaming,dryness can be relieved.Using modern science and technology to conduct preliminary research on the processed product of steamed Citri reticulatae pericarpium,establish the best processing parameters of steamed Citri reticulatae pericarpium,study its quality control methods,and explore its pharmacological effects and the processing mechanism of alleviating dryness after steaming.Methods:1.It has used single factor combined with Box-Behnken response surface method to get the best processing parameters of steamed Citri reticulatae pericarpium.2.It has used Fourier transform infrared spectrometer and GC-MS technology to analyze the composition changes of Citri reticulatae pericarpium before and after steaming.3.By studying the properties,microscopic and thin layer identification,moisture,alcohol/water extract,ash and aflatoxin,volatile oil content and the content of the three main flavonoids of steamed tangerine peel,the quality control method of steamed Citri reticulatae pericarpium was preliminarily established.4.Established a rat model of "spleen-deficiency diarrhea" and a dryness model to explore the pharmacological effects and dryness side effects of Citri reticulatae pericarpium before and after steaming.5.The real-time fluorescent quantitative PCR method was used to detect the expression of AVPR-V2,PKA,AQP2 m RNA in rat colon tissue,and to explore the mechanism of Citri reticulatae pericarpium regulating water metabolism before and after processing.Results:1.The best process for steaming Citri reticulatae pericarpium was to add 30 m L of water for every 100 g of Citri reticulatae pericarpium,moistened it for 40 minutes, and steamed for 3 hours.2.Infrared spectroscopy detected that there were 19 identical absorption peaks and 2 difference peaks in the wave number range of 4000~400cm-1 for Citri reticulatae pericarpium before and after steaming.After the Citri reticulatae pericarpium was steamed,the total volatile oil content decreased,and 14 new components were detected in the volatile oil components,but 11 components were not detected.3.The properties,microscopic identification,thin-layer identification,water content and aflatoxin of the proposed steamed Citri reticulatae pericarpium should meet the requirements of Citri reticulatae pericarpium.It was recommended that the total ash content in the quality standard of steamed tangerine peel should not exceed 5.0%,the total volatile oil content should not exceed 1.0%,the water and alcohol extract content should not be less than 42.5% and 43.0% respectively,and the content of hesperidin,nobiletin and tangeretin should not be less than 5.0%,0.50% and 0.20% respectively.4.Citri reticulatae pericarpium before and after steaming had the pharmacological effects of regulating qi,invigorating the spleen,drying dampness,and stopping diarrhea;Citri reticulatae pericarpium before steaming had dry side effects,and the dryness was alleviated after steaming.5.The expression of AVPR-V2,PKA,and AQP2 m RNA in rat colon tissues could be increased before and after steaming of Citri reticulatae pericarpium,which affected the water metabolism of rats.Conclusion: This experiment conducted a systematic study on the processing technology and quality control of steamed Citri reticulatae pericarpium,the pharmacological effects of tangerine peels before and after processing,dry side effects and the mechanism of regulating water metabolism,and provided references for the modern production of steamed tangerine peels and the safety of clinical medication. |
