ObjectiveTo compare the effects of acupuncture at Shu-Mu acupoints of the large intestinal meridian and non-meridian-non-acupoint on gastrointestinal transit function,interstitial cells Cajal and autophagy related protein p62 in rats with slow transit constipation,so as to provide certain scientific evidence for the clinical application of acupuncture in the treatment of STC.Methods1.Five commonly used Chinese and English databases were searched,and the animal experimental literatures on acupuncture treatment of constipation were included.Through Excel and Cloud Platform of Ancient and Modern Medical Records,the situation of acupoint selection in acupuncture prescription was analyzed to provide reference for acupoint selection in acupuncture treatment of constipation.so as to provide reference for acupoint selection of acupuncture treatment of constipation related research.2.Forty male SD rats were randomly divided into blank group,model group,ShuMu acupoint group and non-meridian-non-acupoint group,with 10 rats in each group.The model group was given compound diphenoxylate suspension by gavage.The ShuMu acupoint group and the non-meridian-non-acupoint group were intervened by electroacupuncture at Dachangshu,Tianshu and non-meridian-non-acupoints respectively.After the intervention,the gastrointestinal motility indexes,ICC and p62 were observed.Results1.A total of 88 articles were included in recent 10 years.The results showed that the most frequently selected acupoints were Tianshu(66),Shangjuxu(51)and Dachangshu(43);the most frequently selected meridian is the stomach meridian of Foot Yangming;The compatibility of acupoints with high degree of correlation in acupuncture prescription was Tianshu-Dachangshu,Quchi-Shangjuxu,TianshuShangjuxu,Dachangshu-Shangjuxu.2.Effect of acupuncture on gastrointestinal transit function in STC model ratsCompared with the blank group,the first black stool excretion time,small intestine propulsion rate and gastric emptying rate of the model group were significantly longer(p<0.01)and lower(p<0.05);compared with the model group,the first black stool excretion time of the Shu Mu acupoint group was significantly shorter(p<0.05)Compared with non-meridian-non-acupoint group,the first black stool excretion time of Shu-Mu acupoint group was shorter(p< 0.05).3.Effect of acupuncture on colon morphology of STC model rats(1)HE staining: compared with the blank group,in the model group,there was slight inflammatory cell infiltration in the lamina propria of colon tissue,with small focal aggregation of lymphocytes or neutrophils;in the Shu-Mu acupoint group,the submucosa,muscle layer and adventitia were closely connected,without obvious pathological changes;in the non-meridian-non-acupoint group,there was slight inflammatory cell infiltration in the lamina propria,with tight connection between muscle layer and adventitia,with slight and obvious pathological changes.(2)Ultrastructure of colonic ICC: compared with the blank group,the ICC cells in the model group had abnormal structure,cristae broken,dissolved or even disappeared,and obvious autophagy was observed.The structure of ICC cells in Shu-Mu acupoint group was normal,and a small amount of autophagosome and primary lysosome were also observed.4.Effect of acupuncture on autophagy level of colonic ICC in STC model rats(1)Western Blot: compared with the blank group,the expression of c-kit and p62 protein in the colon of rats in the model group was decreased(p< 0.01);compared with the model group,the expression of c-kit and p62 protein in the colon of rats in the ShuMu acupoint group and the non-meridian-non-acupoint group was increased(p< 0.01).(2)Immunofluorescence: compared with the blank group,the content of c-kit +p62(photosynthetic)protein in colon tissue of model group was decreased(p< 0.05).Compared with the model group,the content of c-kit + p62(photosynthetic)protein in colon tissue of rats in Shu-Mu acupoint group and non-meridian-non-acupoint group was significantly increased(p< 0.01).Conclusion1.The gastrointestinal transit function of STC rats was impaired,and the number and structure of colonic ICC were abnormal.2.Acupuncture at Shu-Mu points of large intestine can significantly improve the gastrointestinal transmission dysfunction of STC rats and can promote the recovery of the number and structure of colonic ICC.3.Acupuncture intervention may improve gastrointestinal function of STC rats by up regulating the expression of ICC marker c-kit protein and autophagy related protein p62 and inhibiting excessive autophagy of ICC.
