ObjectivesThyroid cancer is the most common malignant tumor of endocrine system.As a result of multi omics research,our understanding of the molecular pathology of thyroid cancer and its subtypes has been significantly improved.But in recent years,the incidence rate of papillary thyroid cancer has been steadily rising,which has become a problem that plagued many countries.It is urgent to solve the early diagnosis and treatment of papillary thyroid cancer,early intervention and prevention of recurrence in the later stage.Therefore,there is still a long way to go if we want to achieve the goals of early detection,early diagnosis,early treatment and good prognosis of papillary thyroid cancer.Therefore,to explore the pathogenesis of papillary thyroid cancer at the molecular level,so as to find out the changes of key genes in the development and evolution of papillary thyroid cancer and find more effective therapeutic targets,so as to achieve the purpose of early detection,early diagnosis and early treatment of papillary thyroid cancer.The purpose of this experimental study is to explore the expression of lncrna Xist in papillary thyroid cancer,and then systematically analyze its expression significance in papillary thyroid cancer tissues,so as to further determine its regulatory effect and mechanism on thyroid cancer cell proliferation,apoptosis,invasion and metastasis.In order to enrich the research status of lncrna Xist in papillary thyroid cancer at home and abroad,and it is very likely to provide new treatment ideas for the clinical treatment of papillary thyroid cancer in the future.Methods1.Select samples,analyze and screen out the differential lncrna between papillary thyroid cancer tissues and adjacent normal tissues,and clarify the differential expression of lncrna Xist in papillary thyroid cancer tissues and adjacent normal tissues(1)110 papillary thyroid cancer tissue samples and adjacent normal tissue samples obtained in the first Department of general surgery of the Affiliated Central Hospital of Shenyang Medical College from April 2020 to October 2021 were selected,and the clinical data of the patients were completely recorded.All tissue samples were frozen in liquid nitrogen immediately after operation and stored at-80℃ until RNA was further extracted.(2)This study used human lncrna microarray v4 0 chip to analyze the difference of lncrna between papillary thyroid cancer tissue and lateral normal tissue.(3)The cells were cultured routinely,and the QRT PCR experiment was carried out to verify the differential expression of lncrna Xist in papillary thyroid cancer and adjacent tissues.2.To observe the correlation between lncrna Xist and the clinical data of patients with papillary thyroid cancer and to clarify the effect of lncrna Xist on the proliferation and invasion of papillary thyroid cancer cells(1)The clinical data of 110 patients with papillary thyroid cancer were collected,and the correlation between the expression level of lncrna Xist and clinicopathological parameters was analyzed by statistical scheme.(2)Thyroid cancer cells TPC-1 and ftc133 were transfected with siRNA.CCK-8 experiment,Transwell experiment and scratch experiment were carried out to study the effect of lncrna Xist on the proliferation,invasion and metastasis of papillary thyroid cancer.3.The data adopts spss20 0 software for statistical analysis and chi square test for hardy Weinberg balance detection.If P>0.05,it indicates that the gene loci in the population selected in this study comply with Hardy Weinberg balance.Chi square test was used to calculate the p value of the distribution of various factors in the case group and the control group,and analyze whether there is any difference.Unconditional logistic regression model was used to remove other external factors,and then the risk of genotypes and alleles in the two groups was calculated.The odds ratio(or)and 95%confidence interval(95%CI)of relative risk were used to analyze and compare the frequencies of genotypes and alleles between the two groups,so as to estimate whether the gene locus is associated with the incidence of thyroid cancer.All statistical results obtained through data analysis showed that the difference was statistically significant(P<0.05).Results1.Lncrna chip screened 12 cases of high expression and low expression standard lncrna in papillary thyroid cancer and adjacent tissues respectively.Volcanic map compared different groups of lncrna and found out the differentially expressed gene lncrna Xi st.2.The results of lncrna chip screening showed that the level of lncrna Xist in papillary thyroid cancer group was significantly higher than that in the control group(P<0.05).3.Between the expression level of LncRNA Xist and the clinicopathological data of patients with papillary thyroid cancer,it was found that the high expression of LncRNA Xist in papillary thyroid cancer was correlated with the presence or absence of lymph node metastasis,TNM stage and degree of differentiation(P<0.01),but not with the age and sex of patients(P>0.05).4.Compared with NC group and blank group,the cell proliferation of TPC-1 and ftc133 cells in Si lncrna Xist group decreased significantly at 0h,24h,48h and 72h(P<0.05).5.The number of TPC-1 and ftc133 cells invaded by NC group and blank group after 24 hours was 182.32±21.12179.45±19.38 respectively,while the number of TPC-1 and ftc133 cells invaded by Si lncrna Xist group was 91.34±13.54 and 94.22±16.21 respectively(P<0.05).6.Compared with NC group and blank group,TPC-1 and ftc133 cells in Si lncrna Xist group healed slowly with the extension of time after 24 hours(P<0.05).Conclusions1.Lncrna Xist is up-regulated in papillary thyroid cancer and correlated with the degree of differentiation,TNM stage and lymph node metastasis,but not with the patient’s age and gender.2.Lncrna Xist is a key regulatory factor to promote the proliferation,invasion and migration of papillary thyroid cancer。...
