Myricaria germanica auct.non Linn.Desv as the Tibetan medicine has the effects of expeling wind to relieve exterior syndrome,relieving cough and detoxification.It is often used to treat rheumatoid arthritis,rheumatoid arthritis and other diseases.At present,the research of M.germanica is mainly focused on chemical composition and pharmacological action,and there is a lack of research on quality evaluation and resource utilization.In this paper,the components of M.germanica were analyzed by UPLC-Q-TOF-MS/MS,which were determined by database and other papers comparison.Network pharmacology was used to study the "component-target-pathway" relationship of M.germanica,to explore the mechanism of anti rheumatoid arthritis,and to determine its quality markers combined with the effectiveness and measurability of chemical components.HPLC was used to determine the content of quality markers and evaluate the quality of M.germanica from different producing areas,so as to provide a theoretical basis for its subsequent quality control and resource utilization.The results are as follows:1.Based on UPLC-Q-TOF-MS/MS technology,this study analyzed the chemical components of M.germanica.Through public database and other papers comparison,47 chemical components were determined,including 19 flavonoids,20 phenolic acids,5 triterpenoids and 3 other compounds.2.Used network pharmacology and molecular docking to explore the anti rheumatoid arthritis mechanism of M.germanica.It was found that it mainly includes41 targets,37 pathways and 28 active components.Among the 41 targets,TNF,VEGFA,ALB,MMP2,atk1 and other targets had higher degree values;The 37 pathways mainly involve Pathways in cancer,TNF signaling pathway,PI3K-Akt signaling pathwayand Hepatitis B;Among the 28 active components,five active components: apigenin,isoquercetin,quercetin,kaempferol and isorhamnetin are closely related to the core targets and important pathways.Through molecular docking verification with the top five key genes: AKT1,VEGFA,TNF,ALB and EGFR,it was found that the five active components have a good combination with the key targets.Combined with the effectiveness and measurability of chemical components,these five components were determined as the quality markers of M.germanica.3.The contents of five quality markers in M.germanica were determined by HPLC.Based on its total contents,combined with principal component analysis and cluster analysis,the quality differences of 30 batches of M.germanica in Qinghai were comprehensively evaluated.The results showed that the content of isoquercetin was the highest,followed by quercetin,and apigenin was the lowest.The best quality of M.germanica was produced in Yushu and guoluongbu in Qinghai,followed was Datong County and Menyuan County.The correlation analysis between the contents of five quality markers and spatial vector showed that quercetin and isoquercetin were significantly positively correlated with altitude(P<0.05);The correlation analysis among the five quality markers showed that isoquercetin was positively correlated with quercetin(P<0.01),and kaempferol was positively correlated with isorhamnetin(P<0.01).
