Research backgroundColorectal cancer(CRC)is one of the most common malignant tumors in the world that threaten human life and health,and its morbidity and mortality increase year by year.Although colorectal cancer has achieved certain results through surgery,chemotherapy,radiation therapy and targeted therapy in recent years,the 5-year survival rate of advanced colorectal cancer is only about 50%,and the prognosis is poor.Therefore,it is particularly important to find new targeted molecules for the diagnosis and treatment of colorectal cancer.Multiple members of A Disintegrin And Metalloprotease ADAM family play key roles in cancer progression by driving multiple processes including angiogenesis,adhesion,cell-to-cell And cell-to-matrix interactions,migration,proliferation,And tumor invasion.ADAM12(A Disintegrin And Metalloprotease ADAM-12)is one of the important members of ADAM family,located on chromosome 10q26.Two subtypes of ADAM12 are produced by splicting variants with different subcellular localization And biological functions.It mainly includes two subtypes: membrane-anchored stereotyped ADAM12(ADAM12-L)and secreted ADAM12(ADAM12-S).Normally,ADAM12 is highly expressed in human full-term placenta.ADAM12 was found to be abnormally high in a variety of solid malignancies,including colorectal cancer.However,there are few studies on ADAM12 in colorectal cancer.To explore the significance of ADAM12 in the occurrence and development of colorectal cancer,and provide theoretical support for subsequent basic and clinical studies.ObjectiveTo explore the expression level of A Disintegrin And metalloprotease-12(ADAM12)in colorectal cancer And its relationship with clinicopathological features of colorectal cancer patients.Method(1)In this study,bioinformatics(multiple online databases)was used to retrieve the expression of ADAM12 in multiple solid tumors and colorectal cancer.(2)This study collected tumor and adjacent tissue samples from 83 patients who underwent surgical treatment from April 2020 to February 2021 in the Department of Gastrointestinal Surgery of Chaohu Hospital affiliated to Anhui Medical University.(3)Immunohistochemistry was used to detect the expression level of ADAM12 protein in cancer tissues and adjacent tissues of colorectal cancer patients.(4)To analyze the expression level of ADAM12 in colorectal cancer and its relationship with clinicopathological features.Results(1)Analysis of TIMER2.0 online databases showed that ADAM12 protein expression was up-regulated in a variety of solid tumors,including colorectal cancer.(2)The GEPIA and GEO databases showed that ADAM12 protein was abnormally high in colorectal cancer compared with normal tissues.(3)Among 83 patients with colorectal cancer in this study,There were56 cases(67.47%)with high expression of ADAM12 protein,27 cases(32.53%)with low expression of ADAM12 protein,13 cases(15.66%)with high expression of ADAM12 protein,and 70 cases(84.34%)with low expression of ADAM12 protein in adjacent tissues,indicating statistically significant differences(χ2 = 45.86 p < 0.0001).(4)The expression level of ADAM12 protein in clinical specimens of colorectal cancer patients and the gender of the patients(p=0.657 χ2 =0.197),age(p = 0.427 χ2 = 0.632),tumor location(p = 0.345 χ2= 0.890)and tumor size(p= 0.906 χ2 = 0.014),degree of differentiation(p= 0.654 χ2=0.201),vascular invasion(p= 0.921 χ2 = 0.010)and nerve invasion(p=0.758χ2=0.095)(p>0.05).The depth of tumor invasion(p=0.003 χ2=8.555),lymph node metastasis(p=0.016 χ2=5.821),TNM stage(p=0.037 χ2=4.329)was closely related to the abnormal high expression of ADAM12 protein,and the difference was statistically significant.Conclusion(1)Bioinformatics(several online databases)suggested that ADAM12 was abnormally high in colorectal cancer(2)ADAM12 is abnormally high in clinical tissue samples of colorectal cancer patients.(3)Highly expressed ADAM12 may play a carcinogenic role in the occurrence and development of colorectal cancer and is closely related to the invasion and metastasis of colorectal cancer(4)ADAM12 may be a potential molecular marker for targeted therapy of colorectal cancer.
