| Objective:To study the protective effect of alcohol-water extract of Periplaueta Americana（APA,Kangfuxin Ye Oral Liquid）on the oxidative damage of testicles in rats caused by cyclophosphamide（CP）and its mechanism,and to provide a theoretical basis and anew way of therapy for the prevention and treatment of reproductive system damage caused by cyclophosphamide,a chemotherapeutic drug used in male tumor patients.Methods:Twenty-four sexually mature SPF male rats（Sprague Dawley,SD）of 8week-old were randomly divided into three groups:control group（CG,n=8）,model group（MG,n=8）and treatment group（TG,n=8）,all of which were regularly fed for 5d.TG was then given APA（0.3ml/10g.d）through oral gavage for 5d.After that,MG and TG were intraperitoneally injected with CP（75mg/kg.d）for 7d,and CG with the same amount of normal saline（NS）.After finishing the animal model preparation,TG was administrated with APA（0.3ml/10g.d）while CG and MG were given NS both through oral gavage for 30d.All rats were then put to death after 24h of fasting.Finally,the body,testis and epididymis weight of rats in all groups were recorded to calculate the organ coefficient of testes and epididymis;the FSH and LH level in the serum of the rats were measured using ELISA;the SOD,MDA and T level in testicular homogenate were also tested;and the damage of seminiferous epithelium of HE-stained testes tissue slice was observed.Results:APA could slow the drop of rats’ body wight,organ coefficient of testes and epididymis induced by CP,and improve their mental state,fur gloss and appetite for food and water.Though partially damaged,the seminiferous epithelium of rats in TG was there to be found through electron microscope,suggesting the protect effect of APA on the epithelium of seminiferous tubules in testicles,hence the ability to produce sperms.Almost no intact seminiferous epithelium was observed in MG,while that was complete in CG as seminiferous tubules filled with mature sperms.In terms of hormone test,FSH and LH levels in MG and TG were evidently higher,compared with in CG（P<0.05）,and the increase in MG was larger.The test results of SOD,MDA and T showed that SOD level dropped in TG and MG,compared with in CG（P<0.05）,while MDA jumped significantly,implying that CP-induced oxidative testicular damage happened to rats’testes in TG and MG.The damage in MG was more evident as the administration of APA protected testicular tissues of rats in TG.T level decreased in MG and TG（P<0.05）,compared with in CG,and much more so in MG.The above results suggest that CP cause oxidative testicular damage probably by breaking the balance between reactive oxygen species and antioxidants in rats,leading to a large accumulation of oxygen free radicals in testicular tissues and lipid peroxidation.That would harm the seminiferous epithelium then further cause spermatogenesis dysfunction,Therefore,a drop in T level and an increase in FSH and LH could be observed.Conclusion:1.APA（Kangfuxin Ye）helps reverse spermatogenic dysfunction in rats triggered by CP-induced oxidative testicular damage.Its mechanism might be related to the antioxidant effect of APA,which reduces hormones,such as FSH and LH in serum,increases MDA in testicular tissues and SOD,then finally restores T level.2.CP,being used as a chemotherapeutic drug,harms the reproductive system of male rats.But the damaged spermatogenic function could be restored through APA antioxidant treatment.3.A regular use of antioxidant drugs（APA）before antitumor therapies might help reduce the oxidative damage of chemotherapeutic drugs to various body systems by improving the body’s own antioxidant capacity.This experiment proves the protective effect of APA on the reproductive system. |
