| Objective We simulate the short time electric thermal damage by creating a rat thermal uterine model to investigate effect of thermal injury to uterus contraction and the mechanism of contraction function variation.Materials and methodsThe virgin rat uterus scalded with 85℃normalsaline for about 90 seconds were used as experimental groups,and the normal uterus as control groups.We compared the contractility induced by 96 m M KCL and oxytocin of the two groups.The expression and activity of contraction associated proteins and m RNA were detected by western blot and real time q PCR analysis.Results(1)Thermal injury uteri after one week did not affect the tissue contractility.(2)Thermal injury sheds the endometrium,and the repair finished in a week.(3)Thermal injury influenced the expression of contraction-associated proteins.the expression of OTR protein decreased significantly but MMP2,MMP-9,and CX-43 increased significantly.(4)Thermal significantly increased the sensitivity of uterus to progesterone and increased the expression of PR-A and TREK-1 which is related to the effect of progesterone.ConclusionsThermal injury had no influence on the contractility of uterus,but significantly increase the sensitivity of myometrium to progesterone.The effect depends on the expression of diverse contraction associated proteins. |
