| Objective: To investigate the effects of Fbxl10 on proliferation,migration and invasion of lung adenocarcinoma cells and its potential mechanism.Methods: Lung adenocarcinoma cell line H1299 and A549 were cultured in vitro and set up blank group(Ctrol group,untreated cells),negative control group(NC group,negative control of Fbxl10),overexpression group(Fbxl10OE group,overexpression of Fbxl10OE)and interfering group(si Fbxl10 group,small interfering RNA of Fbxl10),all were transfected into lung adenocarcinoma cell line H1299(or A549)by liposome method.The effects of Fbxl10 gene on the proliferation of lung adenocarcinoma cell line H1299 were detected by clonal formation assay and MTT assay.The effects of Fbxl10 gene on migration and invasion ability of lung adenocarcinoma cell line H1299(or A549)were examined by cell wound healing experiment and Transwell migration and invasion experiments.The RT-PCR and Western blotting were used to detect the m RNA levels and related protein expressions of Fbxl10 and PI3 K transfected lung cancer cell line.Results: The lung adenocarcinoma cell line H1299(or A549)with Fbxl10 overe xpression and knockdown expression was successfully constructed;cell cloning as say,MTT assay,cell scratch healing assay and Transwell migration and invasion assay were performed,In lung adenocarcinoma cell line H1299(or A549),ove rexpressing Fbxl10,promoted the proliferation,migration of lung cancer cells wit h NC group.(P< 0.05);The results of RT-PCR showed that,when Fbxl10 is o verexpressed,the expression of Fbxl10 m RNA and PI3 K m RNA was higher than that of NC group(P< 0.05);Western blotting showed that,when Fbxl10 is ov erexpressed,the expression of Fbxl10 protein and PI3 K protein was also signific antly higher than that of NC group,difference was statistically significant(P< 0.05).Conclusions: In lung adenocarcinoma cells,there is a close relationship between Fbxl10 and PI3 K,and the high expression of Fbxl10 is also accompanied by the high expression of PI3 K.Overexpression of Fbxl10 could promote the proliferation,migration and invasion of lung adenocarcinoma cells;knockdown its expression can inhibit the proliferation,migration and invasion of lung adenocarcinoma cells. |
