| Kruppel-like factors(KLFs)are a highly conserved zinc finger protein family,involving in various physiological processes such as early embryogenesis,organogenesis and disease formation in mammals.Zinc finger protein 267(ZNF267),as one member of this family,can regulate the proliferation,differentiation,stress reactions,and other processes of cell metabolisms,and is closely related to the occurrences of many diseases,such as Alzheimer’s disease,leukemia,pancreatic cancer,liver cancer,diffuse large B-cell lymphoma,etc.Information given in the biological information databases(GEPIA,UALCAN and CCLE)show that the expression of ZNF267 in breast cancer cells and tissues is higher than that in normal cells and their adjacent normal tissues,and the survival rate of patients with high expression is lower.With respect to the function of ZNF267 in the processes of growth and metastasis of breast cancer cell,related reports have not been associated yet.On the basis of overexpressing ZNF267 in breast cancer cells,this study aims to explore the relationship between ZNF267 and breast cancer cells metastasis.The results can be outlined as follows:1.After cloning the CDS sequences of ZNF267 successfully,the p EB-3×flagZNF267 lentivirus overexpression plasmids had been constructed.Then the p EB-3×flag-GP and p EB-3×flag-ZNF267 plasmid lentiviruses had been packaged,and at last transfected into breast cancer cells(SUM159 and MCF-7).QRT-PCR and Western Blot results showed that ZNF267 overexpressed breast cancer cell lines(SUM159-ZNF267 and MCF-7-ZNF267)had been successfully constructed.2.MTT and cell clone formation experiments showed that the overexpressed ZNF267 can enhance the proliferation activity and tumorigenicity of breast cancer cells.3.Wound healing and Transwell migration experiments showed that the overexpressed ZNF267 can significantly promote the migration of breast cancer cells.4.Transwell invasion assay showed that the overexpressed ZNF267 can enhance the invasive ability of breast cancer cells.5.After transcriptome sequencing of breast cancer cells(SUM159-ZNF267/SUM159-GP and MCF-7-ZNF267/MCF-7-GP),243 and 1225 differentially expressed genes had been identified,respectively.Among them,13 genes are shared by the two cells,involving SLC6A12,DHRS2,GALNT9,S100A9,OLFM1,ACTL10,TMEM8 B,FER1L4,GPNMB,CAPN12,VLDLR,STS and RAB3 B.GO functional annotation and KEGG pathway enrichment analysis had been performed.And after the overexpression of ZNF267,the metabolic pathways that enriched differentially expressed genes were mainly involved in cell adhesion molecule pathway,c AMP signaling pathway,GABAergic synaptic pathway,Rap1 signaling pathway,and cholesterol metabolism pathway,PI3K-AKT signaling pathway,etc.All these results indicate that ZNF267 affects breast cancer cell metastasis via participating in multiple signaling pathways.In conclusion,the overexpression of ZNF267 can promote breast cancer cell metastasis.This study provides new idea for molecular targeted therapy of breast cancer. |
