| Objective: To understand the effects of edaravone on the expression of different subtypes of superoxide dismutase(SOD)in the kidney of rats with intermittent hypoxia,to explore the protective effect of edaravone on the kidney of intermittent hypoxia,and to provide new ideas and theoretical basis for the clinical prevention and treatment of sleep apnea hypopnea related kidney injury.Methods: 36 male SD rats aged 4 weeks were purchased from the animal experi ment center of Lanzhou University and placed in an indoor environment with appropri ate temperature and humidity.The rats were fed with ordinary feed to the age of 8 wee ks.The 36 male rats were randomly divided into 9 rats in each group,including norm oxia control group(group A),intermittent hypoxia group(group B),intermittent hypo xia and edaravone intervention group(Group C),intermittent hypoxia and normal sali ne group(Group D).The rats of group B,group C and group D were placed in the exp erimental cabin respectively,and group A was placed in the indoor environment to fee d without restriction and move freely.Group B,group C and group D were successive ly injected with compressed nitrogen,compressed oxygen and compressed air.The nit rogen pipe was opened in the first 30 s to keep the oxygen concentration in the cabin at6-7% during the rapid hypoxia period and maintain the hypoxia for 10s;We controlle d the experimental equipment to inject compressed oxygen for 20 s and recovered the oxygen concentration into the test cabin,The rapid hypoxia period lasted for 60 s,so t hat the oxygen concentration remained at 20-21%.A total of 120 s was required for a c ycle.We monitored the gas concentration in the cabin and adjusted the gas flow rate in time to make the cabin reach the set oxygen concentration.The modeling time was 8hours per day for 8 weeks.Except for the modeling time,the rats were kept in the ind oor environment at other times.Eight weeks after the establishment of intermittent hyp oxia animal model,the kidney specimens of rats in each group were stained with HE,PAS and Masson.The pathological changes of kidney were observed under light micr oscope,and the pathological damage of kidney was scored.The complete glomerular area of renal tissue stained with PAS and was collected under high-power microscope,and the glomerular area was measured by image analysis software.The contents of S OD1,SOD2 and SOD3 in rat kidney were detected by Elisa.The protein expression o f SOD1,SOD2 and SOD3 was observed by immunohistochemistry,and the correlation between glomerular area and the expression of SOD1,SOD2 and SOD3 was statistic ally analyzed.Result: 1.Compared with group A,SOD1 in group B,group C and group D decreased(P < 0.05);Compared with group B,SOD1 in group C increased(P < 0.05);Compared with group C,SOD1 in group D decreased(P < 0.05).The above data were statistically significant.Compared with group A,SOD2 decreased in group B(P <0.05);Compared with group B and group D,SOD2 in group C changed little(P >0.05);Compared with group A,SOD3 decreased in group B(P < 0.05);Compared with group B and group D,group C had statistical significance(P < 0.05).2.HE staining was observed under light microscope: The renal tissue structure was intact,the renal capsule and tubular structure were not damaged,and the tubular lumen structure was clear in group A.The renal structure was seriously damaged,glomerular capillary congestion,glomerular swelling,incomplete renal capsule structure,serious injury,narrowing of renal tubular lumen,congestion and a small amount of inflammatory cells in the kidney in group B and D;The renal structure,glomeruli and tubules were slightly damaged in group C.(2)It can be seen that the mucus protein secreted by the kidneys in group B and D is the most,the brush edge structure of renal tubular epithelium is incomplete,and the capillary loop collapses;Group A and group C secreted the least mucus protein.In group C,the glomerular basement membrane was slightly thickened and the glomerulus was swollen in PAS staining.(3)Masson staining showed that the content of renal fiber in group A and C was less,that in group C was the least,and that in group B and D was more.(4)Correlation analysis showed that glomerular area was negatively correlated with SOD1(r =-0.485,P < 0.001),SOD2(r =-0.341,P < 0.05)and SOD3(r =-0.445,P < 0.05).Conclusion: 1.Intermittent hypoxia can reduce the expression of SOD1,SOD2 and SOD3.After edaravone intervention,the expression of SOD1 and SOD3 increases,but the expression of SOD2 hardly changes.2.Intermittent hypoxia can lead to renal tissue damage.Edaravone has a protective effect on renal damage caused by intermittent hypoxia.3.Correlation analysis showed that the glomerular area was negatively correlated with SOD1,SOD2 and SOD3.That is the smaller the values of SOD1,SOD2 and SOD3,the larger the glomerular area and the heavier the renal pathological damage. |
