Objective: To investigate the effect of one bead on the activity of NLRP3 inflammatory body on the learning and memory ability of aging rats constructed by D-galactose and its mechanism.Methods: 50 SD rats were randomly divided into normal group,model group and overhead one bead drug dose group(low dose group,medium dose group and high dose group),with 10 rats in each group.The aging model of rats was established by subcutaneous injection of 100 mg / kg D-gal every day in the model group and each drug dose group;At the same time,the low-dose group,medium dose group and high-dose group were gavaged with 50,100 and 200 mg / kg of the extract of one pearl on the head every day;The normal group and the model group were gavaged with the same volume of double distilled water.The quality of rats was detected and recorded every week,and the model was established for 6 weeks.At the 6th week,Morris water maze test was used to detect the behavior of experimental rats;HE staining was used to detect the morphological changes of hippocampus in experimental rats;The expressions of NLRP3 and caspase-1 in the hippocampus of experimental rats were observed by immunohistochemistry;Western blot was used to observe NLRP3,ASC,caspase-1 and interleukin-1 in the hippocampus of experimental rats β(IL-1β)And the expression of interleukin-18(IL-18).Results: 1 Compared with the normal group,the number of neurons passing through the platform,rpl-1 and vacuolar area in the model group was significantly shorter,and the latency of neurons passing through the target area of rpl-1 and rpl-1 in the model group was significantly shorter than that in the normal group.The protein expression level was significantly up-regulated.2.Compared with the model group,the escape latency time of rats in each drug dose group was significantly reduced,the number of times of crossing the survival platform was more,the residence time in Qie target quadrant was longer,the number of neurons in hippocampus was more,arranged closely,nuclear loss was less,vacuolation was less,and NLRP3,ASC,caspase-1,1l-18 and IL-1β in hippocampus were more The protein expression level was significantly down regulated.Conclusion: one bead on the top of the head can improve the learning and memory of aging rats,and its mechanism may be related to down regulating the expression level of related proteins after NLRP3 inflammatory body activation.
