| Objectives To detect the expression levels of Netrin-1,DCC protein and mRNA in bladder invasive urothelial carcinoma,and to explore the relationship between the gene and the invasion and metastasis of bladder invasive urothelial carcinoma and the relationship between the two genes.method1.Immunohistochemical SP method was used to detect the expression of Netrin-1 and DCC protein in 30 cases of bladder invasive urothelial carcinoma and 30 cases of matched normal bladder mucosa as control;2.The expressions of Netrin-1 and DCC mRNA in 30 cases of bladder invasive urothelial carcinoma were detected by in situ hybridization,and 30 cases of matched normal bladder mucosa were used as control;Spss25.0 Application software for statistical analysis,using Fisher’s exact test,Pearson chi square and Spearman rank correlation test.The inspection level is α= 0.05。result1.In bladder invasive urothelial carcinoma,the positive rate of Netrin-1 protein expression(60.0%)was higher than that of normal bladder mucosa(26.7%),and the positive rate of Netrin-1 mRNA expression(70.0%)was higher than that of normal bladder mucosa(40.0%)(P < 0.01).There was a significant positive correlation between Netrin-1 protein expression and mRNA expression(P < 0.01).2.In bladder invasive urothelial carcinoma,the positive rate of DCC protein expression(16.0%)was lower than that of normal bladder mucosa(80.0%),and the positive rate of DCC protein mRNA expression(23.3%)was lower than that of normal bladder mucosa(73.3%).The difference was statistically significant(P < 0.01),and there was a significant positive correlation between DCC protein expression and mRNA expression(P < 0.01).3.In bladder invasive urothelial carcinoma,the positive expression rate of Netrin-1 protein and mRNA was related to clinical stage,vascular / nerve invasion and lymph node metastasis(P < 0.05).4.In bladder invasive urothelial carcinoma,the positive expression rate of DCC protein and mRNA was related to clinical stage,vascular / nerve invasion and lymph node metastasis(P < 0.05).5.There was a significant negative correlation between Netrin-1 protein expression and DCC protein expression in bladder invasive urothelial carcinoma(P < 0.01).6.There was a significant negative correlation between Netrin-1 mRNA expression and DCC mRNA expression in bladder invasive urothelial carcinoma(P < 0.01).Conclusion the expression of Netrin-1,DCC protein and mRNA is related to the occurrence,invasion and metastasis of bladder invasive urothelial carcinoma.Netrin-1gene is an important promoter of the occurrence and development of bladder invasive urothelial carcinoma,and DCC gene is an important inhibitor of the occurrence and development of bladder invasive urothelial carcinoma,... |
