BackgroundAcute myocardial infarction(AMI)due to coronary artery blockage is currently one of the leading causes of morbidity and mortality in the world.Thrombolytic therapy,interventional therapy,and coronary artery bypass grafting are important methods for timely recovery of blood supply,relief of angina,and reduction of coronary heart disease mortality,but the restoration of blood supply will inevitably cause ischemia reperfusion injury(IRI).The preganglial fibers of the sympathetic nerve that innervate the heart are distributed to the sinoatrial node,the atrioventricular bundle,the atrioventricular node,the atrial muscle,and the ventricular muscle after stellate ganglion.Stellate ganglion block inhibits overactive sympathetic nerves,dilates coronary arteries,and reduces myocardial oxygen consumption.ObjectivesTo investigate whether stellate ganglion block can alleviate myocardial ischemia-reperfusion oxidative stress injury in rats.MethodsThirty SPF adult male SD(Sprague Dawley)rats,were equally divided into three groups according to the random number table method:control group(group C),ischemia-reperfusion group(group R),and stellate ganglion block group(Group S).The left anterior descending coronary artery(LAD)was ligated to create a rat model of myocardial ischemia-reperfusion.In the control group(group C),the silk thread only passed through the left anterior descending coronary artery without ligation;in the ischemia-reperfusion group(group R),the left anterior descending coronary artery was ligated for 30 minutes,and then the blood flow was restored for 120 minutes;The stellate ganglion block group(group S)also ligated the left anterior descending branch of the coronary artery for 30 min,and the ropivacaine planetary ganglion block was used to restore blood supply for 120 min.The heart rate(HR)and mean arterial pressure(MAP)were recorded before surgery(T0),30 minutes after ligation(T1),30 minutes after reperfusion(T2),60 minutes after reperfusion(T3),and 120 minutes after reperfusion(T4).After the experiment,the myocardial injury was reflected by recording the serum troponin I(c Tn I),creatine kinase isoenzyme(CK-MB)and lactate dehydrogenase(LDH)in the three groups of rats.Malondialdehyde(MDA)and superoxide dismutase(SOD)in oxidative stress reflect the level of oxidative stress.Results1.By ligating and reopening the anterior descending branch of the left coronary artery in rats,a model of myocardial ischemia and reperfusion injury in rats was successfully made.2.By comparing the changes of heart rate and blood pressure in three groups of rats at different times,it was found that:There was no statistical difference in blood pressure and heart rate among the three groups at time T0.At time T1,blood pressure and heart rate increased significantly in groups R and S compared with group C(P<0.05).Compared with group C,blood pressure still decreased significantly and heart rate increased significantly,but blood pressure in group S was higher than that in group R(P<0.05),and heart rate in group S was lower than that in group R(P<0.05).With the passage of time,the blood pressure of the rats in the C group did not change much,the blood pressure in the R group gradually decreased,and the blood pressure in the S group became stable after reperfusion.3.The levels of serum troponin I(c Tn I),creatine kinase(CK-MB)and lactate dehydrogenase(LDH)in three groups of rats were higher in group R and group S than in group C(P<0.05).But group S was lower than group R(P<0.05).4.Comparison of serum MDA and SOD levels in three groups of rats,R group,S group MDA level is higher than C group,SOD level is lower than C group,the difference is statistically significant(P<0.05),the myocardial MDA content of S group is lower In the R group,the SOD content was higher than that in the R group(P<0.05).ConclusionStellate ganglion block can improve myocardial perfusion,and has a certain protective effect on myocardial ischemia-reperfusion injury by reducing the level of oxidative stress.
