| BackgroundHepatocellular carcinoma(HCC)is a common malignant tumor with a 5-year survival rate of only 7%.Early onset symptoms of HCC are not obvious,so it is easy to lead to the discovery of advanced stage,delayed diagnosis and treatment.NLRP6 is a member of the NLR family of inflammasome,which can play a number of roles,including maintaining cell integrity,promoting injury healing and inhibiting cellular inflammation.Studies have shown that the inflammasome is highly expressed in liver disease,but the correlation between NLRP6 and HCC is unclear.Therefore,it is of great significance to search for tumor markers or targeted molecules for the early diagnosis and treatment of HCC.ObjectiveTo detect the activation levels of inflammasome NLRP6 and its downstream inflammatory factors Caspase-1,IL-1β,IL-18 in HCC tissues,and to analyze the correlation between inflammasome NLRP6 and its downstream inflammatory factors Caspase-1,IL-1β,IL-18 and HCC.To clarify the value of the combined detection of inflammasome and its downstream inflammatory factors in the early diagnosis of HCC,the evaluation of postoperative efficacy and prognosis of HCC patients,and further improve the positive rate of HCC diagnosis.MethodThis study intends to collect 52 hepatocellular carcinoma patients in Change People’s Hospital from January 2018 to June 2021 as the research object,and select 55 non-liver cancer patients during the same period as the control group.The m RNA expression levels of NLRP6 inflammasome and its downstream inflammatory factors Caspase-1,IL-1β and IL-18 in peripheral blood and liver tissues of patients were detected by real-time fluorescence quantitative PCR.The protein expression levels of NLRP6 inflammasome and its downstream inflammatory factors Caspase-1,IL-1β and IL-18 in peripheral blood of the two groups were detected by ELISA.The protein expression levels of NLRP6 inflammasome and its downstream inflammatory factors Caspase-1,IL-1β and IL-18 in liver tissues of patients were detected by Western Blot.Furthermore,various liver function biochemical indexes in peripheral blood of the two groups were detected,and the relationship between the expression level of NLRP6 inflammosome in peripheral blood and inflammatory factors,as well as the relationship between the m RNA expression level of these indexes and various liver function biochemical indexes were analyzed.Result1.The m RNA and protein expression levels of NLRP6,Caspase-1,IL-1β and IL-18 in peripheral blood of patients with HCC were higher than those of control group(P<0.05).2.The m RNA and protein expression levels of NLRP6,Caspase-1 and IL-1β in liver tissues of patients with HCC were lower than those of control group(P<0.01).3.The m RNA and protein expression levels of IL-18 in peripheral blood and liver tissues of patients with HCC were higher than those in the control group,which had a certain correlation with the biochemical indexes of liver function.ConclusionThe expression of NLRP6 inflammasome was related to the related activities in HCC.IL-18 may be an important indicator for predicting the prognosis of the disease and evaluating the severity of the disease,but its specific mechanism and possible signaling pathway need further study. |
