BackgroundNasal bone fracture is the interruption of nasal bone continuity asa result of external forces acting on the Nasal bone.Clinically,nasal bone fracture is often manifested as nasal bleeding,local pain,nasal obstruction,appearance collapse malformation,which seriously affects the normal nasal ventilation and appearance of patients.If the treatment is not timely,delayed healing of fracture or bone defect,external nasal malformation,etc.,is a common disease in otorhinolaryngology.Recent studies have found that growth factors released by platelets during fracture healing can trigger angiogenesis,activate proliferation and chemotaxis of mesenchymal cells,osteoblasts and chondrocytes,and accelerate bone healing.At present,studies on the promotion of platelet-rich plasma PRP in the healing of bone defects of long bone fractures have been reported at home and abroad,but no relevant studies have been found on the promotion of PRP in the healing of nasal bone defects.In this regard,we tried to make a rabbit nasal bone defect model.To investigate the promoting effect of platelet-rich plasma(PRP)on nasal bone defect healing in rabbits.ObjectiveTo investigate the therapeutic effect of platelet-rich plasma on bone healing of nasal bone defect model in rabbits,and to provide experimental basis for clinical application.Methods24 adult new zealand white rabbits,weighing 2.5~3.0 kg,all male,were selected to make cylindrical nasal bone defects with a diameter of 5mm and a depth of 2.5mm.They were randomly divided into PRP group(group A)and blank group(group B),with 12 rabbits in each group.Gross observation,histological staining(HE staining,Masson staining and fluorescence staining),CT value,col-1 and alkaline phosphatase levels were measured at 4,8 and 12 weeks postoperatively.SPSS26.0 software was used to process the data.Results1.The rabbit nasal bone defect model was successfully made;2.The results of histological staining showed that there was no infection or death in each group after operation.The bone defects in group A and group B were healed to varying degrees,and the healing speed of bone defects in group A was stronger than that in group B;3.CT measurement: Postoperative CT finger measurement of group A and Group B showed that group A was significantly higher than group B at 4 weeks,8 weeks and 12 weeks,the difference was statistically significant(P< 0.05);4.type ⅰ collagen: group A was significantly higher than group B at 4 weeks,8 weeks and 12 weeks,the difference was statistically significant(P< 0.05);5.The results of bone alkaline phosphatase determination showed that the level of group A was higher than that of group B at all postoperative periods,and the difference was statistically significant(P< 0.05).ConclusionPlatelet-rich plasma(PRP)has a strong promoting effect on the repair of nasal bone defects in rabbits.The mechanism may be that the tissue density,type ⅰ collagen and BALP content of newborn bone tissue can promote the repair and healing of nasal bone defects.
