In recent years,the use of polarized light to characterize the structural properties of biological tissues has become an emerging direction.The heterogeneous and highly scattering properties of biological tissue result in a large degree of optical depolarization and are therefore prone to loss of polarization signal information.It is one of the main research directions to develop high-sensitivity measurement and effective data analysis methods to solve these problem.The main work of this paper is based on the BX51 microscope,which adds a patterned polarization modulation microscopic imaging system,Firstly,the expression of the extinction angle and phase delay amount of the sample and the expression of the Stokes parameter of the outgoing light are theoretically derived.Subsequently,imaging experiments were performed on several histopathological sections,and applied the obtained Extinction angle(?),scattering depolarization(Idp)and phase delay(?sin??)polarization parameters and stokes parameters for imaging analysis and calculation to analyze the imaging characteristics of the lesion tissue;By comparison,it can be found that the linear phase delay parameter value and the scattering depolarization parameter value of the diseased tissue sample are small overall,and has a strong extinction;Then,based on the polarization microscopy imaging system,the polarized camera is used instead of the ordinary camera to perform the Mueller matrix detection of the sample.Using horizontal,vertical,45 degree Linearly polarized light and a right-handed circularly polarized light transmitting source illuminate the biological tissue sample,Polarization camera is used to obtain the polarization parameter map of the light emitted under the four light source conditions,then the Mueller matrix of biological tissue was calculated by Matlab image processing.It was found that a variety of structural information and optical properties of the sample can be obtained by imaging with a polarized Mueller microscope.Finally,the in-vivo imaging and detection of polarized light is studied,and a liquid crystal fast phase modulation polarized light imaging optical path is built.and the Stokes parameters are theoretically calculated by using the Mueller matrix,and which is applied to human skin detection,then the imaging results are analyzed,and the relationship between the intensity of the scattered light of the skin and the polarization angle of the incident light is obtained,and the theoretical fitting is performed.The findings are in good agreement with the actual measurement results,which verifies the feasibility and correctness of the system.
