| ObjectiveTo observe the effects of Jiangya Tongluo Formula on blood pressure and renal function,and the regulation of ADM and its receptors in rats with hypertensive nephrosclerosis.The study aimed to explore the pathway and mechanism in protecting kidney which was affected by the formula.Thurs,it will provide new therapeutic target and new ideas for clinical treatment and drug screening approaches.MethodsSixteen-week-old Wistar Kyoto rats were used as control group.Even-aged spontaneously hypertensive rats were randomly divided into 5 groups:model group,valsartan group,Jiangya Tongluo Formula groups which included high dose,median dose and low dose ones.All the rats were gavaged respectively.The tail arterial pressure was detected before gavage,after treated 2 weeks,4 weeks,6 weeks and 8 weeks respectively.Urine samples were collected for detecting 24h urinary protein content,urinary P2-MG.After treated 4 weeks and 8 weeks,serum creatinine,blood urea nitrogen levels,serum β2-MG and content of ADM in kidney and plasma were estimated after each group was sacrificed.HE staining was made after paraffin section to be observed renal pathological morphological changes after removing one side of the kidney.The protein expressions of ADM receptor systerm which included CRLR,RAMP2,RAMP3 were detected by western blotting.ResultsAfter treated 4 weeks and 8 weeks,the rats tail arterial pressure,serum creatinine level,blood urea nitrogen content,24h urinary protein content,urinary β2-MG in treated groups decreased and the content of ADM in kidney increased significantly than those in model group(P<0.05,P<0.01)and the content of ADM in kidney was increased significantly in treat groups(P<0.05,P<0.01).The blood pressure of rats with hypertensive nephrosclerosis increased with the dose of Jiangya Tongluo Formula.And compared to control group,the content of ADM in plasma was increased significantly in model group and two treat groups after 4 weeks,but decreased at the end of treated 8 weeks,and there were no statistically significance between treated groups and models at the two time points.The protein expression of ADM and its receptors in the kidney were up-regulated in the treatment groups compared to control group.The glomerular ischemia and tubular sclerosis were improved which contributed to the recovery of renal structure and function.ConclusionWe choosed the model of rats with hypertensive nephrosclerosis,treated with Jiangyatongluo Formula and the Valsartan in the study.Analying the experiment results,the conclusions were got as follow:1.Jiangyatongluo Formula could reduce the blood pressure of rats with hypertensive nephrosclerosis,alleviating glomerular sclerosis.It protected kidney and blood vessel from hypertension.2.Jiangyatongluo Formula could reduce the serum creatinine and urea nitrogen of rats with hypertensive nephrosclerosis,contributing to the protecting renal function.3.Jiangyatongluo Formula could reduce 24-hour proteinuria and urine β2 microglobulin of rats with hypertensive nephrosclerosis,alleviating the function of glomerular filtration and tubule reabsorption.4.Jiangyatongluo Formula could increase the content of ADM in kidney and up-regulate the protein expressions of ADM and its receptors.The protective function of JYTL may be through up-regulation local ADM level in kidney regardless of circulating ADM.This may be one of the machenisms of hypertensive nephrosclerosis. |
