Experimental Study On Elastic Fibers, MMP-7 And TIMP-1 In The Filtering Tract Scar Tissue Of Qingguang'an After Anti-glaucoma Surgery

Object:By observing the effect of scarring area of filtration canal after glaucoma surgery through the four QingGuangAn effective groups and QingGuangAn granules,to discuss and compare their mechanism of action on scarring area of filtration canal.Method:Selected 48 healthy adult NewZealand long ears white rabbits,they were outbred,male or female,the average weight were 1.5～2.0kg.Then the rabbits were divided into 8 groups randomly with 6 in each group.These groups were:group A was blank group;groupB was model group;groupC was MMCgroup;groupD、E、F、G were the four QingGuangAn effective groups;groupH was QingGuangAn ganules group.Every group did the trabeculectomy and Iris root excision surgery except the group A,groupC used MMC during the operation.All model groups were given medicine in the next day,and measured intraocular pressure,observed the anterior segment of eyes and filtration bleb.After using four weeks medicine,all rabbits were executed by air injection at ear vein,the whole organization of filtration canal were taken and deposited in the fixative.The elastic fiber were detected with Masson,the expression of MNP-7and TIMP-1 in scarring area were detected with IHC.Results:Compared with the Preoperative basic IOP and the second day,the seventh day,the fourteenth day,the twentyeighth day postoperative IOP of groupC,groupE and groupH,we found that the IOP of three group rise up slower than other groups,and they were kept the lowest data at the twentyeighth day,compared with the other groups showed statistic significance.GroupC,H,E could formed functional bleb and maintained at the twentyeighth day,groupD,F,G could maintained parts of functional bleb,however some model eyes had already became the no functional bleb.Bleb of groupB began to disappeared until filled with tissues around at the twentyeighth day.GroupC came out a special model eye that its conjunctiva necrosis,and the conjunctiva,cornea,atria,crystalline lens and the fundus oculi of each operative group eyes had no serious complications.The area density of elastic fiber:compared with operative groups and blank group,P<0.05,showed statictis significance,comared with groupC,H,E,P>0.05,showed no difference,comparing groupH and groupE,P>0.05,showed no difference.Other intergroup comparation showed statistic significance.The AO of MMP-7,TEMP-1:the expression showed statistic significance except groupC and groupH,groupC and groupE,groupH and groupE,groupB and group G,groupF and groupG,their P>0.05,showed no difference.Conclusion:The scarring area of filtration canal after glaucoma surgery is the major reason which lead to the failure of surgery.Qing Guang An effective group 2,Qing Guang An granules and MMC could reduced the scar tissue by restrained the elastic fiber,MMP-7 and TIMP-1.By observing the experimental results that both QingGuangAn effective group2 and QingGuangAn granules could restrained the scarring area of filtration canal,the effects were unbiased,QingGuangAn granules group is better than effective group2.
