Objective:The vessel restenosis model was established by denuding aortic endothelium with domestic balloon catheter in rats.Extracellular matrix deposition and inflammatory response as the main target to study the effect of Astragalus combined with Angelica on intimal hyperplasia.To determine whether Astragalus combined with Angelica can inhibit ECM deposition and inflammatory response.Methods:Sprague-Dawley rats were divided into different groups by baseline method:Astragalus-Angelica different proportion group(Crude drug 3.9g/kg)?Astragalus group(2.17g/kg)?Angelica group(1.08g/kg)?Positive drug atorvastatin control group(0.1g/kg)and Sham operation group.Each group had 8 rats.A model of intimal hyperplasia of thoracoabdominal aorta was established by balloon catheter injury in rat vascular endothelium.Then the thoracoabdominal aorta was taken out after administration of Astragalus and Angelica with different ratios for 14 days.The expression of main components FN,Co1-? and ICAM-1,VCAM-1 and IL-1? of ECM were determined by immunohistochemistry.The expression of MMP-9 and TIMP-1 in ECM was determined by Western blot.The expression of IL-1?,TNF-? and MCP-1 in plasma was determined by ELISA.Result:The expression of ECM-related factors in the proliferative endometrium of each group:Compared with the sham group,the expression of Co1-?,FN and MMP-9 in the proliferative endometrium of the model group was significantly increased(P<0.01).Compared with the model group,the expression intensity of Co1-?,FN and MMP-9 in Angelica group,Astragalus-Angelica 1:1 group,Atorvastatin group were significantly lower(P<0.01-P<0.05).The expression of Col-? and MMP-9 in Astragalus-Angelica 5:1 group was significantly lower than that in model group(P<0.05).The expression of MMP-9 in Astragalus group was significantly lower than that in model group(P<0.05).There was no significant difference in the expression intensity of Col-? and FN between the Astragalus group and the model group(P>0.05).There was no significant difference in the expression intensity of FN between the Astragalus-Angelica 5:1 group and the model group(P>0.05).There was no significant difference in the expression intensity of TIMP between each group(P>0.05).Comparison of localized inflammatory factors in each group:Compared with the sham group,the expression of ICAM-1,VCAM-1 and IL-1? in the model group was significantly increased(P<0.01).Compared with the model group,the expression of ICAM-1,VCAM-1 and IL-1? in Angelica group,Astragalus group,Astragalus-Angelica 1:1 group,Astragalus-Angelica 5:1 group and Atorvastatin group were significantly higher(P<0.01-P<0.05).The expression of IL-1? in Astragalus group was significantly lower than that of the model group(P<0.05).There was no significant difference in the expression of ICAM-1 and VCAM-1 between the Astragalus group and the model group(P>0.05).The levels of plasma inflammatory factors IL-1?,TNF-? and MCP-1 in each group were compared:Compared with the sham group,the levels of IL-1?,TNF-? and MCP-1 in the model group were significantly increased(P<0.01).Compared with the model group,the levels of IL-1?,TNF-? and MCP-1 in Atorvastatin group were significantly lower than those in the model group(P<0.01).There was no significant difference between the other drug groups and the model group(P>0.05).Conclusion:Angelica and Astragalus-Angelica 1:1 combination can inhibit the deposition of ECM in the proliferative endometrium,inhibit the migration and proliferation of VSMC,and reduce the local inflammatory response.Thereby inhibiting the role of inhibition of intimal hyperplasia.In the compatibility of Astragalus-Angelica,Angelica is the main effect of the drug,Astragalus-Angelica 1:1 compatibility is stronger than 5:1.
