Objective:Selecting the etiology composite modeling method to est ablish models of HP associated gastritis spleen-stomach dampness-heat syndrome,evaluating synthetically the value of this method in the pro cess of establishing models of HP associated gastritis spleen-stomach dampness-heat syndrome,and to clarify the relationship between gastri c mucosal injury of models of HP associated gastritis spleen-stomach dampness-heat syndrome and FoxO signal pathway that regulating apo ptosis.Methods:60 BALB/c mice were randomly divided into 5 groups,com pounding control group,internal wet HP group,external wet HP group,si mple HP group and complex etiology group.Twelve mice in each gro up that half male and half female.Selecting the complex etiology mod eling method(fat and sweet food + hygrothermal environment + HP)t o establish models of HP associated gastritis spleen-stomach dampness-heat syndrome.Selecting two mice in each group(one male and one fe male)to evaluate whether modeling is successful,after the finish of m odeling.Indicator analysis including:general state performance of mice;HSP70、AQP4;pathological morphology and degree of inflammation of gastric mucosa tissue;evaluation of gastric motility and secretory funct ion(MTL and GAS cootent in the serum);evaluation of apoptosis of ga stric mucosa cells;detection of substances related to signal pathway.Results:1.Compared with normal control group,the spleen-stomach dampness-h eat syndrome manifestations(delayed reaction、lethargy and idleness、l anguid、decline in diet and drinking water、Loose stool)of different degrees and change of anal Temperature increase were observed in ea ch model group.Comparing with other model groups,the symptoms of complex etiology group were more prominent.2.Compared with normal control group,the expression levels of HSP70 and AQP4,and the secretion of MTL increased in each model gro up.However the secretion of GAS is decreased.Among them,the mos t obvious change was in the complex etiology group.3.There was no obvious inflammatory reaction in gastric mucosa of normal control group,gastric mucosal inflammation in model group cha nged to different degrees.The inflammatory reaction was more obvious in the complex etiology group.4.Detection of apoptotic rate of gastric mucosa cells:The positive rat e of apoptotic cells increased in model group,And the highest positiv e rate was found in the complex etiology group.5.The expression levels of PTEN and FoxO3 a in each model group w ere higher than those in the normal control group.But the expression l evel of PI3 K and Akt was lower than that of normal control group.T he most significant change was in the complex etiology group.Conclusion:Complex etiology method is superior to other methods.This method is more in line with the clinical process of Spleen-stoma ch dampness-heat syndrome.And plays an important role in the establi shment of HAG spleen-stomach damp-heat syndrome model in mice w ith combination of disease and syndrome.The apoptotic rate in HAG model of Spleen-stomach damp-heat syndrome is increased,and the apo ptotic mechanism is related to PTEN/PI3K/Akt/FoxO signal pathway,by activating its pathway to regulate apoptosis.
