Objective: To study the effect of the mechanism of Mieyou Decoction in the expressions of P27、Fasl and Bim in the H.Pylori associated gastritis mice with spleen and stomach damp-heat syndrome.Methods: 1)60 BALB/c mice were randomly divided into six groups including the control group,Complex cause model group,the low-does Mieyou Decoction group,the medium-does Mieyou Decoction group the high-does Mieyou Decoction group,the Gastric quadruple therapy group,Male and female are kept separately.2)The mice of H.Pylori associated gastritis mice of spleen and stomach damp-heat syndrome model were established by composite factor including the fatness food,the hygropyrexia environment and Helicobacter pylori.Observe the symptoms of the model animals(nose,pill or constipation,burnout),water intake,diet,body weight,rectal temperature,hair color and hair smoothness.3)After the success of modeling,Thecontrol group and the Complex cause model group were treated with distilled water,and the other groups were given the corresponding drugs for 14 days.4)continuous administration two week,Rapid urokinase assay for HP planting,The pathological changes of gastric mucosa and the degree of inflammation were observed under HE staining.The apoptosis rate of gastric mucosa was detected by TUNEL method.the P27、Fasl and Bim expression was detectd by the Western-blot and RTPCR method.Results: 1)Before the administration,two mice in each group were tested for rapid urokinase,the mice in the other groups were positive for rapid urokinase except for the control group.2)The pathological changes of gastric mucosa and the degree of inflammation were observed by HE staining under light microscope.The model group was significantly different from the control group.The inflammation in the treatment group was alleviated except for the low-dose Mieyou decoction group.3)TUNEL method was used to detect the apoptotic rate of gastric mucosa cells.The apoptotic cells in the Complex cause model group were significantly increased compared with the normal group.The treatment group improved except for the lowdose Mieyou decoction group.4)Compared with the control group,expressions of P27 were significantly decreased,Fasl and Bim were significantly higher in the Complex cause model group(P<0.01);Compared with the Complex cause model group,expressions of P27 were significantly higher,Fasl and Bim were significantly decreased in the medium-does Mieyou Decoction group,the high-does Mieyou Decoction group,the Gastric quadruple therapy group(P<0.01);Compared with the high-does Mieyou Decoction group,the change about expressions of P27、Fasl and Bim were no significant difference in the medium-does Mieyou Decoction group(P>0.05);Compared with the high、 medium does-Mieyou Decoction group,the change about expressions of P27、Fasl and Bim were significant in the Gastric quadruple therapy group(P<0.01);Compared with the control group,expressions of P27、Fasl and Bim were no significant difference in the low-does Mieyou Decoction group(P> 0.05).Conclusion: The high、medium does-Mieyou Decoction may play a role in treatment of H.Pylori associated Gastritis with Spleen and Damp-heat Syndrome through down Inhibition of apoptosis by Regulation of expression of apoptotic factors P27、 Fasl and Bim downstream of Fox O signaling pathway.
