The thesis mainly focused on the chemical constituents isolated from the roots of Rhododendron Micranthum and their bioactivities.R.Micranthum belongs to the genus of Rhododdendron in the family of Ericacea,which is wide-spread in regions of North China,Northeast China and Henan,Shandong,Hunan,Hubei,and Sichuan provinces of China.In China,the branch and leaves of were used as a traditional Chinese medicine for the treatment of chronic tracheitis,dysmenorrhea,fracture,hypertension,puerperal arthrodynia and rheumatoid arthritis.In order to provide the chemical evidence for the rational application of this Chinese folk medicine,a lot of phytochemistry investigations on the leaves and branches of R.micranthum have been proceeded,and revealed the presence of lignans,sesquiterpenes,coumarins,triterpenes,and diterpenoids.But there has no report of the constituents of the roots of R.micranthum.Therefore,encouraged by the enriched structures and significant activities of grayanoids,the roots of R.micranthum(17 kg)were collected from Yiyuan Shandong Province in August 2014.In the thesis,54 compounds were isolated from the root of R.micranthum,including nineteen grayanane diterpeniods(1-19),seven triterpenes(20-26),two steroids(27-28),five sesquiterpenes(29-33),one iridoid glycoside(34),two monoterpenes(35-36),eight lignans(37-44),one flavone(45),one lactam(48),five phenolic acids(49-53),one benzoate(54),and two other types compounds(46-47).Among them,three new compounds(1-3)were acquired.On the basis of spectral analysis,54 compounds were elucidated as 16?-acetoxy grayanotoxin ?(1),3?,6?,16?-trihydroxy-14?-acetoxy-grayan-1(5),10(20)-diene(2),16?-acetoxy rhodomollein ???(3),grayanotoxin ?(4),grayanotoxin ?(5),grayanotoxin ?(6),rhodomicranol D(7),grayanotoxin ?(8),grayanotoxin ?(9),grayanotoxin ??(10),16-acetylgrayanotoxin ?(11),1-epi-grayanotoxin ?(12),iso-grayanotoxin ?(13),grayanotoin ?(14),grayanotoxin ?(15),grayanotoxin ?(16),grayanotoxin ?(17),rhodomicranol C(18),16-anhydrograyanoroxin ?(19),4-epi-barbinervic acid(20),2?,3?,19?,23-tetrahydroxyurs-12-en-28-oic acid(21),2?,3?,23-trihydroxyurs-12-en-28-oic acid(22),2?,3?,23-trihydroxy-olean-12-en-28-oic acid(23),3?,23-dihydroxy-olean-12-en-28-oic acid(24),betulinic acid(26),7?-hydroxysitosterol(27),7?-hydroxysitosterol(28),anhuienosol(29),hedytriol(30),pterocarptriol(31),(1S,4S,5R,7S,10S)-trihydroxyeudesmane(32),1?,5?,7?-guaiane-4?,10?,11-triol(33),monotropein methyl ester(34),paeoveitol B(35),(1S,2S,4R)-2-hydroxy-1,8-cineole-?-D-glucopyranoside(36),lyoniside(37),(-)-(7R,8S,8'S)-lyoniresiol(38),2,3-bis[(4-hydroxy-3,5-dimethoxyphenyl)-methyl]-1,4-butanediol(39),(+)-isolariciresinol(40),ssioriside(41),2,3-bis[(4-hydroxy-3,5-dimethoxyphenyl)-methyl]-1,4-butanediol(42),nudiposide(43),cedrusin(44),taxifolin(45);(2RS)-2-(hydroxy-methyl)-1,4-dioxacyclodotriacontan-5-one(46);octanedioic acid(47);1,6-hexanolactam(48);2,3-dihydroxy-3-(4-hydroxy-3,5-dimethoxypheny)-1-propanone(49);vanillic acid(50);syringic acid(51);p-coumaric acid(52);2-(3',4'-dihydroxyphenyl)-1,3-benzodioxole-5-aldehyde(53);dibutylphthalate(54).An acetic acid-induced writhing test in mice was proceeded to evaluate the antinociceptive activities of compounds 1-3,5-6,9-14 and 16.Compared to vehicle-injected mice,compounds 1,5,6,14 and 16 showed significant antinociceptive effects with writhe inhibition rates of 45.8%-64.2%at a dose of 0.1 mg/kg and compounds 10,12 and 13 showed significant antinociceptive effects with writhe inhibition rates of 33.9%-64.8%at a dose of 5 mg/kg.Compound 2 showed potent antinociceptive effects with writhe inhibition rates of 86.1%and 54.7%at doses of 8 mg/kg and 0.8 mg/kg,respectively.
