| Objective: In the early stage of this project,big data analysis was carried out on the relationship between Bushen Huoxue Recipe and postmenopausal osteoporosis from the network pharmacology level,to predict the therapeutic effect of Bushen Huoxue Recipe on postmenopausal osteoporosis and the core target of the drug’s effect on the disease(IL-6,TNF-α),and observe the effects of this formula on the serum IL-6,TNF-α and bone mineral density of the proximal femur in ovariectomized rats through animal experiments in the later period,verify and explore the effect of Bushen Huoxue Recipe on postmenopausal bone The curative effect of porosity and whether the adjustment of IL-6 and TNF-α levels in serum can be used to intervene in the formation of osteoporosis,through big data analysis combined with experiments,provides relevant evidence for the prevention and treatment of postmenopausal osteoporosis in traditional Chinese medicine.Methods: In the early stage,a number of databases and software were used to screen out the core target with research value.In the later stage,animal experiments were carried out.The animals were bred adaptively for 1 week before the animals were modeled.After 1 week,32 adult female SD rats were weighed and combined.Stratified,divided into 4 groups by random number table method: blank group,estrogen group,traditional Chinese medicine group,8 animals in each group.In the traditional Chinese medicine group,2.5g of Bushen Huoxue Recipe was administered every two months after the model was made;the estrogen group was given 1.5ml of the prepared conjugated estrogen solution every day two months after the model was made.The blank group and the model group were in the two models.After months,he was given an equal volume of normal saline by gavage every day.The gavage of each group lasted three months in total.Three months later,the rats in each group were sacrificed.After execution,the serum IL-6 and TNF-α levels were detected by enzyme-linked immunoassay.The bone density of the proximal right femur of the rats was detected by dual-energy X-ray,and the data were obtained and statistically analyzed.Results:1.Bushen Huoxue Recipe can intervene in multiple ways of postmenopausal osteoporosis through multiple targets,which has certain therapeutic significance,and it is likely to play a key role in its treatment through core targets with high Degree values such as IL-6 and TNF.2.Bone mineral density: Compared with the blank group,the bone mineral density of the rats in the traditional Chinese medicine group,the estrogen group and the model group all decreased(P<0.01);but the bone mineral density of the rats in the estrogen group and the traditional Chinese medicine group was significantly higher In the model group(P<0.01);the traditional Chinese medicine group was compared with the estrogen group,the P value was 0.722,and there was no significant difference in bone density,suggesting that the two have the same curative effect.3.IL-6 and TNF-α: Serum IL-6 and TNF-α levels in model group were significantly higher than those in blank group(P< 0.01);the serum IL-6 of rats in the Chinese medicine group and estrogen group TNF-αis lower than the model group(P < 0.01);while the comparison between the blank group,traditional Chinese medicine group and estrogen group is not statistically significant(P> 0.05)Conclusion:1.Bushen Huoxue Recipe can reduce the levels of serum IL-6 and TNF-α in ovariectomized rats.2.Bushen Huoxue Recipe regulates the imbalance of bone metabolism,increases bone mineralization,improves bone,and has a certain therapeutic effect on postmenopausal osteoporosis.3.Bone mineral density was negatively correlated with serum IL-6and TNF-α levels.4.The experimental results are consistent with the prediction and analysis of the Bushen Huoxue Recipe-target gene(IL-6,TNF-α)-PMOP by network pharmacology,that is,Bushen Huoxue Recipe has a certain curative effect on PMOP,and part of the effect is through the regulation of IL-6 in serum,TNF-α level is achieved. |
