Objective: In this study,by observing the effect of "Xing Nao Kai Qiao" acupuncture on the cerebral infarction area and related targets in the brain tissue of MCAO model rats,and to analyze the network target topology to explore the possible mechanism of acupuncture treatment of cerebral infarction The size and interaction with each target.Methods: 200 SPF SD rats were randomly divided into 50 normal group,50 sham operation group,50 model group and 50 acupuncture group.Acupuncture was performed in the acupuncture group and the sham operation group,and the other two groups were only tied without needles.The four groups were operated for 20 minutes each time,twice a day.Brain tissues were taken from each group at five time points,1d,3d,5d,7d,and14 d after operation.4 rats were randomly selected at each time point to detect the area of cerebral infarction,and 6 rats were randomly selected to detect inflammation-related targets(TNF-α,IL-1β,IL-6,NF-κB,ICAM-1,P38MAPK),cell necrosis and apoptosis-related targets(Caspase-3,Bcl-2,AIF,MMP-9,MMP-2,ERK1),oxidation Related targets(SOD,HO-1),thrombus-related targets(P-selectin),repair and regeneration-related targets(BDNF,TGF-β,VEGFR2).The mathematical model constructed by the law of mass action is used to calculate the size of the 18 targets and their relationship.Results:1.Cerebral infarction area: The cerebral infarction area of the acupuncture group was significantly smaller than the model group 14 days after the operation(P<0.05),and there was no difference in the acupuncture group compared with the model group at the rest of the time.2.Inflammation-related target detection:(1)The content of TNF-α in brain tissue:there was no difference in each group on the 1st postoperative day;the content of the acupuncture group was significantly lower than that of the model group at 3d,5d,7d,and14 d after operation(P<0.05).(2)The content of IL-1β,IL-6,NF-κB,ICAM-1,P38 MAPK in brain tissue: There was no difference in the acupuncture group at 1d,3d,5d,7d after the operation of these 5 targets compared with the model group;14d after operation The content of the acupuncture group was significantly lower than that of the model group(P<0.05).3.Detection of cell necrosis and apoptosis-related targets:(1)The content of Caspase-3 in brain tissue: There is no difference in the acupuncture group at 1d,3d,5d,7d after operation compared with the model group;the content of the acupuncture group at 14 d after operation is more significant than that of the model group Decrease(P<0.01).(2)Bcl-2,AIF,MMP-9,ERK1 content in brain tissue: There was no difference in the acupuncture group 1d,3d,5d,7d,14 d after the operation of these 4 targets compared with the model group.(3)The content of MMP-2 in brain tissue: 1d,3d,5d,7d after operation,the model group had no difference compared with the acupuncture group;14d after operation,the content of the acupuncture group was significantly lower than that of the model group(P<0.05).4.Oxidation-related target detection:(1)SOD content in brain tissue: 1d,3d,5d,7d,14 d after operation,the acupuncture group has no difference compared with the model group.(2)The content of HO-1 in brain tissue: the model group was no different from the acupuncture group at 1d,3d,5d,7d after operation;the content of the acupuncture group was significantly higher than that of the model group at 14 d after operation(P<0.01).5.Thrombus-related target detection:(1)The content of P-selectin in brain tissue: 1d,3d,5d,7d after operation,the acupuncture group had no difference compared with the model group;14d after operation,the content of the acupuncture group was significantly lower than that of the model group(P< 0.05).6.Detection of targets related to repair and regeneration:(1)The content of BDNF and TGF-β in brain tissue: There was no difference in the acupuncture group at 1d,3d,5d,7d,14 d after the operation of these 2 targets compared with the model group.(2)The content of VEGFR2 in brain tissue: There was no difference in the acupuncture group at1 d,3d,5d,7d after operation compared with the model group;the content of the acupuncture group was significantly higher than the model group at 14 d after operation(P<0.05).7.Network target topology analysis: In the network relationship between the targets,NF-κB and Caspase-3 have the strongest positive effect,and BDNF and NF-κB have the strongest negative effect;18 targets have the same position and are evenly dispersed;Among them,ICAM-1,P38 MAPK,MMP-2,Bcl-2,MMP-9,AIF,BDNF,IL-1β,ERK1 play a greater role.Conclusion:1."Xing Nao Kai Qiao" acupuncture method can effectively reduce the cerebral infarct area of rats;2."Xing Nao Kai Qiao" acupuncture can down-regulate the content of TNF-α,IL-1β,IL-6,NF-κB,ICAM-1,P38 MAPK in the brain tissue of MCAO model rats and inhibit inflammatory reaction damage;3."Xingnao Kaiqiao" acupuncture can down-regulate the content of Caspase-3 and MMP-2 in brain tissue of MCAO model rats and inhibit cell necrosis and apoptosis;4."Xing Nao Kai Qiao" acupuncture can down-regulate the content of P-selectin in brain tissue of MCAO model rats and inhibit the formation of thrombus;5."Xing Nao Kai Qiao" acupuncture can increase the content of VEGFR2 in brain tissue of MCAO model rats and improve the blood supply of cerebral infarction area;6."Xing Nao Kai Qiao" acupuncture can increase the content of HO-1 in brain tissue of MCAO model rats to resist free radical damage;7.NF-κB and Caspase-3 have the strongest positive effects in the network,and BDNF and NF-κB have the strongest negative effects;8.The 18 targets have the same status in the network,each is independent,and there is no obvious key target;9.Among the 18 targets,ICAM-1,P38 MAPK,MMP-2,Bcl-2,MMP-9,AIF,BDNF,IL-1β and ERK1 play a greater role in the network.
