Objective:To observe the effect of Xinjia Xiaoke decoction on glucose and lipid metabolism,and the expression of pancreatic duodenal homeobox factor-1(PDX-1)in rats with type 2 diabetes mellitus(T2DM)induced by streptozotocin(STZ),to explore the possible mechanism of Xinjia Xiaoke decoction in the treatment of type 2 diabetes mellitus.Methods:The rats model of type 2 diabetes mellitus(T2DM)were established by intraperitoneal injection of streptozotocin(STZ)combined with high glucose and high fat diet for 4 weeks.T2DM model rats were randomly divided into three groups,model group,Xinjia Xiaoke decoction group and glibenclamide group,with 10rats in each groups.Another 10 SD rats were used as control group.The rats in Xinjia Xiaoke decoction group were given Xinjia Xiaoke formula 15.3 g·kg-1·d-1by gavage,The rats in the glibenclamide group was given glibenclamide 0.23 mg·kg-1·d-1by gavage.And the rats in the model group and the normal group were given the same amount of normal saline under the same conditions.At the end of 4,6,8 and 10 weeks,the body weight and FBG were measured by glucose oxidase method.Glucose tolerance test(OGTT)was performed at the end of 10 weeks.At the end of 4 and 10weeks,the FINS level and HOMA-IR were detected by enzyme-linked immunosorbent assay(ELISA).At the end of 10 weeks,the rats were killed and the blood was collected from abdominal aorta.The glycosylated serum protein level was detected by enzyme-linked immunosorbent assay(ELISA).The levels of TC and TG were detected by automatic biochemical analyzer.The expression of PDX-1 protein and m RNA in pancreatic tissue were detected by Western blotting and real-time PCR.Results:Before administration(4 weeks),compared with the normal group,there was no significant difference in the body weight level of the model group(P>0.05),the levels of FBG,GSP and insulin resistance indexin the model group were significantly increased(P<0.01).Compared with the model group,there were no significant differences in the FBG,body weight,FINS and insulin resistance index of the Xinjia Xiaoke decoction group and glibenclamide group(P>0.05).After administration(4 weeks),compared with the normal group,the FBG level of the rats in model group was significantly increased at the end of 6,8 and 10 weeks(P<0.01),and the weight of the the rats in model group was significantly decreased at the end of6,8 and 10 weeks(P<0.01).The levels of FINS,insulin resistance index,GSP,TC,TG and OGTT blood glucose levels were significantly increased(P<0.01).The expression levels of PDX-1 protein and m RNA in pancreatic tissue were significantly decreased(P<0.01).Compared with the model group,the level of FBG in Xinjia Xiaoke decoction group was decreased at the end of 6,8 and 10 weeks(P<0.01),and the level of body weight was increased at the end of 6,8 and 10 weeks(P<0.01);the fasting insulin,insulin resistance index,glycosylated serum protein,triglyceride,total cholesterol and OGTT blood glucose levels decreased at different degrees(P<0.05 or P<0.01).The levels of PDX-1 protein and m RNA expression in pancreatic tissue were significantly increased(P<0.01).The levels of body weight and FBG in glibenclamide group were significantly increased at the end of 6,8 and 10weeks(P<0.01).The levels of FINS,insulin resistance index,glycosylated serum protein,TC,TG and OGTT blood glucose were decreased in varying degrees(P<0.05or P<0.01).There was no significant difference in the expression of PDX-1 protein and m RNA(P>0.05).Conclusion:1.Xinjiaxiaoke decoction can significantly reduce the levels of FBG,FINS,glycosylated serum protein,TC and TG in T2DM rats.Xinjiaxiaoke decoction can increase glucose tolerance,recover pancreatic injury and improve insulin resistance.2.The mechanism may be related to the up regulation of PDX-1 protein and m RNA expression in pancreatic tissue.
