Objective:To establish a rat model of kidney-yang deficiency type osteoporosis,then observe the effect of Jingang Pills on OPG,RANKL protein expression and its key signals TAB2,AP-1,and explore the medicine of Jingang Pills for preventing and treating kidney-yang deficiency type postmenopausal osteoporosis effective basis and mechanism of action.Methods:Seventy SD rats were divided into blank group(Nt C),sham operation group(SHAM),and model group(OVX)according to the random number table method.Ovariectomy combined with hydrocortisone intramuscular injection modeling was used to establish kidney yang deficient ovariectomized rat model.After 8weeks of model building,3 rats were randomly selected from each group,and the femoral bone density and the rats’ behavioral observations were used to verify the model.After successful model verification,the model rats were divided into model group,estradiol valerate group,and low,medium,and high doses of King Kong pills.Each administration group was given the corresponding drugs once a day for 12 weeks,sham operation The same volume of distilled water was also given to the stomach in the group,the blank group,and the model group.At 6weeks and 12 weeks after administration,ELISA method was used to detect serum OPG,RANK,RANKL,TAB2 and AP-1 content,Western Blot method was used to detect rat femur OPG,RANKL,TAB2,AP-1 expression;12 weeks after administration,using HE staining method for bone histomorphometric observation.Results:(1)The bone mineral density of model rats was significantly lower than that of the blank group at 8 weeks after model building(P<0.05),which was successfully verified with the model of kidney-yang deficiency in rats.(2)At 6weeks and 12 weeks after administration: Compared with the blank group,there was no significant difference on OPG,RANKL,TAB2 and AP-1 in the sham operation group(P>0.05);In the model group,the OPG content of rats was significantly reduced,and the content of RANKL,TAB2,AP-1 was significantly increased,and the difference was statistically significant(P<0.05).The OPG content of rats in the administration group decreased,and the content of RANKL,TAB2,AP-1 increased,the difference was statistically significant(P<0.05);Compared with the model group,the OPG content of rats in the sham operation group and each administration group was significantly increased,and the content of RANKL,TAB2 and AP-1 was significantly decreased(P<0.05);Compared with the high-dose pill group,the OPG,RANKL,TAB2 and AP-1content of rats in the estradiol valerate group,the Jingang pill medium-dose group,and the Jingang pill low-dose group did not improve significantly,and the difference was statistically significant(P<0.05);Compared with the estradiol valerate group,there was no significant difference in OPG,RANKL,TAB2 and AP-1 content between the medium and low dose groups of Jingang Pill(P>0.05);Between the medium and low dose groups of Jingang Pill The difference was not statistically significant(P>0.05).(4)The pathological results showed that Jingang pills can improve the morphological structure of bone trabeculae and adjust the arrangement of bone cells.Conclusion:Jingang Pills can affect the expression of TAB2 and AP-1 by regulating the OPG/RANK/RANKL signaling pathway,thereby preventing and treating postmenopausal osteoporosis due to deficiency of kidney-yang.
