| The transmission of information between neurons is accomplished through the release and reabsorption of neurotransmitters.Vesicles are the places where neurotransmitters are stored and released,and are the basic unit of chemical information transmission between neurons.According to reports,the abnormal storage and release of neurotransmitters were related to some neurological diseases,such as Parkinson’s disease and Alzheimer’s disease.Therefore,studying the impact of potential drugs on the storage and release of neurotransmitters is of great significance to the development of drugs for related neurological diseases.Glochidion eriocarpum Champ.is Glochidion of Euphorbiaceae plants and Panax ginseng C.A.is Panax L.of Araliaceae plants,previous studies have shown that ginsenoside Rg1 and polysaccharides of Glochidion eriocarpum have neuroprotective effects.In this paper,a single-cell electrochemical analysis method for neurotransmitter storage and release was established,and we studied the effects of ginsenoside Rg1 and polysaccharides of Glochidion eriocarpum Champ.on the storage and release of neurotransmitters in PC 12 cells,which provided theoretical basis for the development and utilization of medicinal plant resources.The main research results are as follows:1.Establishment of a single-cell electrochemical analysis method for neurotransmitter storage and release.A single-cell electrochemical analysis platform was built using carbon fiber microelectrodes,inverted microscopes and HEKA amplifiers.Using the electrochemical analysis platform combined with the intracellular vesicle impact electrochemical cytometry method and single cell amperometric method,the quantitative analysis of the storage and release of neurotransmitters in the vesicles in a single PC 12 cell was realized,which confirmed the feasibility of the electrochemical analysis platform and method.2.The effect of ginsenoside Rg1 on the storage and release of neurotransmitters.Ginsenoside Rg1 is an important medicinal plant component in ginseng,and it is reported to have a good neuroprotective effect.However,it has not been reported whether it has an effect on the storage of neurotransmitters in vesicles and the release of neurotransmitters during exocytosis.This paper uses the established electrochemical analysis method to study its effect on the storage and release of neurotransmitters in PC12 cell vesicles.The results showed that the storage of neurotransmitter in a single vesicle of the Rg1 group(112950±8460,N=35)was not significantly different from that of the control group(98100±9990,N=30),indicating that ginsenoside Rg1 had an effect on PC 12 cell vesicles.The content of the content has no significant effect.The results of the exocytosis experiment showed that compared with the control group,Nevents and Imax were significantly increased(p<0.01),and the trise,t1/2 and tfall of the Rg1 group were significantly reduced(p<0.05),indicating that ginsenoside Rg1 has an effect on PC 12 The exocytosis of cell vesicles has a significant impact.The specific manifestation is that Rg1 treatment increases the frequency of exocytosis events,increases the size of the exocytosis hole,and greatly shortens the residence time of the exocytosis hole.The results of tfall and release ratio show that ginsenoside Rg1 makes the neurotransmitter tend to be in partial release mode.3.The effect of polysaccharides from Glochidion eriocarpum Champ.on the storage and release of neurotransmitters.There are few reports on the chemical information transmission of SPZch between neurons.This paper used the established platform and methods to study its effects on the storage and release of neurotransmitters in PC 12 cell vesicles.The experimental results of neurotransmitter storage showed that compared with the control group(98100±9990,N=30),the SPZch group(150260±11370,N=25)had a significant increase in neurotransmitter storage in a single vesicle(p<0.001),indicating that SPZch significantly increased the neurotransmitter storage in PC 12 cell vesicles.The results of exocytosis showed that compared with the control group,Imax and Nevents were significantly increased in SPZch group(p<0.0001),and trise,t1/2 and tfall were significantly decreased(p<0.01),indicating that SPZch increased the occurrence of exocytosis frequency,increased the size of the exocytosis hole and greatly shorten the residence time of the exocytosis hole.The results of the release ratio show that SPZch makes the neurotransmitter exocytosis more inclined to a partial release mode. |
