Study On Preparation Technology, Quality Standard And Efficacy Of Shaoyao Gancao Decoction

Purpose:Using this method that central composite design combing with CRITIC toptimize the preparation technology of peony and licorice decoction.It based on the preparation technology to study quality standrad andanalgesic and anti-inflammation effects.The study use modern science tegnologies and methods to study peony and licorice decoction,based on peony and licorice decoction of Zhang zhongjing’s Treatise on Febrile and Miscellaneous Diseases.The study sysnthetically appraises peony and licorice decoction from its finger-print,dose effect relationship and index of content of paeoniflorin,albiflorin,liquiritin,ammonium glycyrrhetate ets and evaluates peony and licorice decoction’s quality to establish bassion of its quality specification study.From dose effect relationship of peony and licorice decoction,the study can know this relationship between different doses of peony and licorice decoction and anti-inflammatoryand analgesic action and lay a foundation of medicine safety.Material and method:1.Using this method that central composite design combing with criteria importance though intercrieria correlation investigated these three factors:extraction time,solid-liquid ratio,extraction times and calculated these four indices of weight coefficient.Using content of paeoniflorin,albiflorin,liquiritin,ammonium glycyrrhetate as index and optimizing the preparation technology of peony and licorice decoction.2.Using HPLC anlyzes this six effective chemical components: aeoniflorin,albiflorin,liquiritin,ammonium glycyrrhetate,liquirtin apioside,isoliquiritin.(1)Using HPLC anlyzes the content of aeoniflorin,albiflorin,liquiritin,ammonium glycyrrhetate in peony and licorice decoction.(2)Using HPLC anlyzes the content of liquirtin apioside,isoliquiritin in peony and licorice decoction.3.Using HPLC establishes the finger-print of peony and licorice decoction.4.This study can know this relationship between different doses of peony and licorice decoction and anti-inflammatoryand analgesic action,and establish dose effect relationship which use hot and cold difference method,acetic acid writhing and albumin-induced paw edema in mouses to investigate the effect of different doses of peony and licorice decoction of anti-inflammatoryand analgesic action.Result:1.The weight coefficients of aeoniflorin,albiflorin,liquiritin,ammonium glycyrrhetate calculated by CRITIC were 0.22、0.18、0.39、0.21.The preparation technology was as follow : the solid-liquid ratio was 1:6,extraction time was three hours,extraction times was three times.2.The experiment takes the method of HPLC to analyze the content of six index components,including the content of paeoniflorin is high in 5.149mg/g～8.997mg/g,the content of isoliquiritin is low in 0.09138 mg/g～0.2282 mg/g.This method is stable and reliable.Each of index components can be tested maximum absorption wavelength.3.Taking ten different batches peony and licorice decoction,this part established the finger-print of peony and licorice decoction.The finger-print of peony and licorice decoction totally calibrated fourteen chromatographic peaks,including six chromatographic peaks from radix paeoniae alba,eight from glycyrrhiza.Results shew that,in this finger-print of peony and licorice decoction,the peak area of effective components were bigger than other components,the peak area of other components were small.4.In the experiments of hot and cold difference method and acetic acid writhing,the high,medium and low doses of peony and licorice decoction had better pesticide effection of analgesic action,however,in the experiment of albumin-induced paw edema,the high,medium of peony and licorice decoction had pesticide effection of anti-inflammatory,and had a dose effect relationship.This experiment shew that the high of peony and licorice decoction had better anti-inflammatoryand analgesic action.Conclusion:From studying peony and licorice decoction,it established quality specification of peony and licorice decoction.One hand the experiment studied quality specification of peony and licorice decoction,on the other hand,it investigated pharmacologica action of peony and licorice decoction from prepartion technology,assaying finger-print and dose effect relationship.So on thia bassion,the study of peony and licorice decoction was sciencal,reasonable and safe,and established a bassion of exploitation and deep investigation.
