Purpose:In this study,acupuncture combined with Xijing Tongmai Decoction was used to treat healthy SD male rats,and then the model of acute myocardial infarction was induced by high fat feeding combined with the injection of isoproterenol hydrochloride.To explore the regulatory effect of acupuncture combined with Xijing Tongmai Decoction pretreatment on the expression of ATP-sensitive potassium ion channel related proteins Kir6.1 and Kir6.2 in rats with acute myocardial infarction,so as to explore the mechanism of acupuncture combined with medicine in improving acute myocardial infarction at the level of ion channel and verify its advantages.Material and method:Healthy male SD rats were randomly divided into 5 groups with10 rats in each group,which were control group,model group,acupuncture and drug combination preconditioning group,Chinese medicine preconditioning group,and western medicine preconditioning group,respectively.After adaptive feeding for a week,the rats were preconditioned.The acupuncture and medicine combination pretreatment group and the traditional Chinese medicine pretreatment group were given by Xijing Tongmai Decoction at8 o ’clock every day,once a day,with 2ml gavage each time.The western medicine pretreatment group was given at the same time every day with the prepared aspirin solvent,with 2ml gavage each time.After intragastric administration,acupuncture and medicine combined pretreatment group selected three points,Neiguan,Xinshu and Zusanli,and used "Huatuo" 0.18×0.25 mm acupuncture needles to obviously stab into the subcutaneous 2mm at each point.After the rats were stabilized,the 6805-D electroacupuncture instrument was applied with a density wave with a frequency of(2-20)Hz.The current intensity was appropriate when the limbs of the rats were slightly tremor and the frequency was consistent with the frequency of electroacupuncture.Acupuncture was performed once a day at 10 o’clock for 15 minutes each time.Both the control group and the model group were wonderstruck given 2ml of normal saline at 8 o ’clock a day.Pretreatment for 2 weeks,during which general feeding,after the end of modeling.The control group was fed with a normal diet(15g/ animal)for 3 weeks,and the other groups were fed with the same amount of high-fat food for 3 weeks under the same conditions,and their electrocardiogram was recorded.The rats were anesthetised by intraperitoneal injection of 10% chloral hydrate(30mg/kg).The rats in the control group were injected with normal saline(85mg/kg)at multiple points at one time(the inner part of the limbs).The rats in the remaining groups were injected with isoproterenol hydrochloride(85mg/kg)in the same way to replicate the animal model,once every 24 hours,twice in total.At the same time,the ECG was observed.After modeling,the mice were sacrificed,samples were collected,and pathological sections of the hearts of each group were observed,and the protein expression levels of ATP-sensitive potassium ion channels(Kir6.1 and Kir6.2)were detected by Western Blot.Results:1.Compared with the control group,the ST segment of model group,acupuncture and drug combination pretreatment group,traditional Chinese medicine pretreatment group and western medicine pretreatment group were significantly elevated;Compared with model group,ST wave increased in acupuncture and drug combination pretreatment group,traditional Chinese medicine pretreatment group and western medicine pretreatment group(P < 0.05),especially in acupuncture and drug combination pretreatment group(P < 0.05),but there was no statistical significance in ECG change in traditional Chinese medicine pretreatment group(P > 0.05).2.The pathological pictures showed that the morphology and structure of the myocardial cells in the control group were normal,the nuclei were ovoid,and there were no fractures,defects or blank areas.The myocardial fibers were arranged neatly and the texture was clear.In the model group,myocardial cells were arranged disorderly,nuclei and myocardial fibers were swollen,and a large number of inflammatory cells were infiltrated.The texture was fuzzy,and fractures,defects and blank areas could be seen.The changes of myocardial cells and myocardial fibers were observed in all the pretreatment groups,but the changes were the least in the acupuncture and drug combination pretreatment group.3.Compared with the control group,the protein expressions of ATP-sensitive potassium ion channel related proteins Kir6.1 and Kir6.2 in model group were increased,and the differences were significant(P < 0.05);Compared with model group,the protein expression levels of Kir6.1 and Kir6.2 in acupuncture and medicine combination preconditioning group,Chinese medicine preconditioning group and western medicine preconditioning group were decreased(P < 0.05).Compared with the acupuncture and medicine combination pretreatment group,the protein expression levels of Kir6.1 and Kir6.2 in TCM pretreatment group and western medicine pretreatment group were significantly decreased(P < 0.05).There was no significant difference in index protein expression between TCM pretreatment group and western medicine pretreatment group(P > 0.05).Conclusion:1.When myocardial infarction occurs in rats,ST segment of ECG will appear elevated.Acupuncture combined with the treatment of Xijing Tongmai Decoction can effectively prevent the changes of ST segment of ECG.The ST segment of ECG in the acupuncture and drug combination pretreatment group was lower than that in the other pretreatment groups.2.The expressions of ATP-sensitive potassium ion channel related proteins Kir6.1 and Kir6.2were normal in the cardiomyocytes of normal rats.When myocardial infarction occurred in rats,the expressions of Kir6.1 and Kir6.2 were increased.The expression levels of Kir6.1 and Kir6.2 were low after pretreatment with acupuncture and drug combination.The expression levels of ATP-sensitive potassium ion channel related proteins(Kir6.1,Kir6.2)in cardiomyocytes were all low after traditional Chinese medicine pretreatment,western medicine pretreatment and acupuncture-drug combination pretreatment,and the index expression levels of acupuncture-drug combination pretreatment group were even lower.3.Traditional Chinese medicine preconditioning,western medicine preconditioning and acupuncture combined with medicine preconditioning had different protective effects on myocardial cells and myocardial fibers,and the combination of acupuncture and medicine had the best protective effect.
