Objective: To study the effect and mechanism of Buyang Huanwu Decoction on pulmonary fibrosis in miceMethods: Using bleomycin-induced pulmonary fibrosis models in mice,they were randomly divided into control group(Con),model group(BLM),Buyang Huanwu Decoction low-dose group,and Buyang Huanwu Decoction medium-dose group,Buyang Huanwu Decoction high-dose group.In the control group,sterile saline(2.5mg/kg)was instilled into the airway,and BLM(2.5 mg/kg)in the airway of the model group and experimental group was instilled to construct a mouse idiopathic pulmonary fibrosis model.All mice were given intragastric treatment on the second day of modeling.Con group and BLM group were given normal saline at 4.5ml/kg,and Buyanghuanwutang low-dose group was given 2.25ml/kg concentrated liquid,medium dose The group was given the concentrated liquid at 4.5ml/kg,and the high-dose group was given the concentrated liquid at 9ml/kg.All mice were administered once a day for 28 consecutive days.Observe the weight,general condition and mortality of the mice.The mice were sacrificed 28 days later,the serum TGF-β1 content was detected by ELISA,the pathological changes of lung tissue were observed by HE staining and Masson staining,and the expression of Smad2,Smad3,α-SMA,Collagen Ⅲ protein in lung tissue was detected by Western blot.Results:(1)Compared with the model group,each dose group of Buyang Huanwu Decoction can improve the general condition and survival rate of IPF mice,but there is no statistical difference between the groups.(2)Compared with the model group,the extracellular matrix and collagen deposition in the lung tissue of the mice in each dose group of Buyang Huanwu Decoction were significantly reduced,and the degree of fibrosis was reduced.With the increase of the drug dose,the extracellular matrix,collagen deposition and fiber The degree of chemistry showed a downward trend.(3)Compared with the model group,the serum TGF-β1 secretion level of mice in each dose group of Buyang Huanwu Decoction decreased significantly,and the expression of Smad2 and Smad3 protein in lung tissue was significantly reduced;as the dose of Buyang Huanwu Decoction increased,TGF-β1,Smad2 and Smad3 gradually decreased,which was negatively correlated with the dosage.Conclusion:Buyang Huanwu Decoction can improve the general condition of pulmonary fibrosis mice and relieve the pathological changes of pulmonary fibrosis in mice.These effects may be through inhibiting the expression of TGF-β/Smads pathway,thereby inhibiting the extracellular matrix(ECM)Deposition plays a role in delaying the progression of pulmonary fibrosis in mice.
