| Object: To observe the effect of Astragalus Polysaccharide Injection on the chemotherapy of Hep G2 tumor bearing mice,and analyze its mechanism.Methods: Using BALB / C nude mice to establish Hep G2 tumor bearing animal model,cisplatin + doxorubicin group(CDDP + ADM group,CA group)was used as positive control to observe the effects of astragalus polysaccharides(APS)and the combination(CA+ APS group)on the health status and tumor growth of tumor bearing mice and tumor inhibition rate were calculated;then the effect of APS on chemotherapy was assessed by peripheral blood and bone marrow examination.Western blot was used to detect the expression of Caspase-3,Caspase-9 and resistance gene MRP1 in tumor tissues of each group,the protein expression among the groups was compared and analyzed,and the mechanism of APS enhancing and reducing toxicity in the chemotherapy of liver cancer was discussed.Results: 1.The synergistic effect of APS on the chemotherapy of liver cancer: the tumor volume and model group of each combination group and CA Group,middle APS group and high APS group were all decreased(P < 0.05).The tumor weight of the three combination groups was significantly lower than that of CA Group(P < 0.05).The tumor inhibition rate of combination group 1 was 50.46%,that of combination group 2 was 68.37%,and that of combination group 3was 62.16%,which were higher than that of CA Group(48.08%).2.The attenuation effect of APS on chemotherapy of liver cancer:the number of peripheral white blood cells and bone marrow cells in the APS group alone compared with the model group,the number of peripheral white blood cells and bone marrow cells in the CA Group and the combination group were decreased(P < 0.05),and the value of the combination group was decreased less than that in the CA Group(P < 0.05).3.Study on the mechanism of APS in the chemotherapy of liver cancer: the expression of Caspase-3 and Caspase-9 in CA Group and three combined drug groups was increased compared with other groups;the expression of MRP1 in CA group increased,and the expression of MRP1 in three combined drug groups was decreased compared with other groups,especially in combination drug group 2.Conclusion: 1.Astragalus polysaccharide injection combined with chemotherapy can improve the tumor inhibition rate of Hep G2,and had synergistic effect on cisplatin + doxorubicin chemotherapy.2.Astragalus polysaccharide injection could reduce the weight,peripheral blood picture and bone marrow inhibition of chemotherapy drugs on Hep G2 tumor bearing mice,and had the effect of reducing toxicity.3.Astragalus polysaccharide injection could inhibit the expression of MRP1 protein,but also up regulate the expression of Caspase-3 and caspase-9,and then promoted the apoptosis of tumor cells.The synergistic and toxic effect of astragalus polysaccharide injection on chemotherapy drugs was closely related to the above mechanism. |
