Study On Preparation Of Ganoderma Lucidum Compound And Its Effect On Sleep Improvement

Insomnia may easily lead to memory recession,decrease immunity and functional failure,and is becoming a common mental illness in the present.In this study,Ganoderma lucidum extracts with water and alcohol extraction were obtained first.Then different G.lucidum compounds were formed and their effects on sleep improvement induced by the combination of pentobarbital sodium were studied.The study provided a scientific basis for the development of functional foods to improve sleep.The main findings were as follows:1.Six groups of medicaments were made by the preparation of G.lucidum extract,preparation of G.lucidum compound and freeze drying:G.lucidum water extract low dose group,G.lucidum water extract high dose group,G.lucidum alcohol extract low dose group,G.lucidum alcohol extract high dose group,G.lucidum compound Ⅰ group(including G.lucidum alcohol extract,Chinese medicine oyster,Semen Ziziphi Spinosae)and G.lucidum compound Ⅱ groups(including G.lucidum water extract,oysters,Semen Ziziphi Spinosae).The six groups of medicaments were subjected to a direct sleep test,a subliminal hypnotic test for pentobarbital,a prolonged sleep test for mice induced by pentobarbital sodium,and a sleep latency test for barbiturates.The results showed that there was no significant change in body weight and no direct sleep effect in each of the drug groups compared with the control mice after continuous administration for 21 days.Compared with the control group,G.lucidum water extract low dose could significant shorten the sleep latency in mice.G.lucidum water extract high dose not only could obviously prolong the sleep time in mice induced by pentobarbital sodium,shorten the sleep latency period,but also prolong the sleep latency period of mice induced by pentobarbital sodium threshold.The high dose of G.lucidum alcohol extract could significantly prolong the sleep duration of the mice induced by pentobarbital sodium threshold and shorten the latency of sleep,but did not prolong the sleep time of the mice.G.lucidum compound(Ⅰ)could significantly prolong the sleep time of mice,shorten the sleep latency,and significantly prolong the sleep duration of mice induced by pentobarbital sodium threshold.The low dose of G.lucidum alcohol extract and G.lucidum compound(Ⅱ)had no obvious effect on the sleep of mice.The comprehensive sleep experiments showed that G.lucidum compound Ⅰ had the best effect on sleep in mice,and this compound was used as a follow-up study.2.After 21 days of continuous administration and 1 hour after the last administration,the mice were sacrificed by cervical dislocation and brain tissues were taken.The brain neurotransmitters and cytokines were measured by enzyme-linked immunosorbent assay.It was found that G.lucidum compound can significantly increase the whole brain in mice 5-Serotonin(5-HT)and dopamine(DA)levels reduce orexin,neuropeptide P(SP),tumor necrosis factor(TNF),nitric oxide(NO),and interleukin-1 in the brain(IL-1)and nitric oxide synthase(NOS)content.The mechanism of action of G.lucidum compound on sleep may be related to the promotion of 5-HT and DA levels of neurotransmitters in the brain and inhibition of Orexin A,SP,and NO levels in the brain.The effects of cytokines IL-1β,TNF-α and NOS on the improvement of sleep need to be further studied.Fluorescence quantitative PCR technique was used to detect the expression of gaba a gene encoding GABA A receptor al subunit in mouse brain.It was found that G.lucidum compound could significantly increase the transcriptional expression of GABAA receptor al subunit gene.It is concluded that G.lucidum compound may improve sleep by promoting the synthesis of GABAA receptor al in mice brain.3.Determination of the-ingredients in G.lucidum compound showed that the polysaccharide,volatile oil and amino acid content of G.lucidum oyster compound were 10.49%,0.95%and 0.89%respectively.In summary,the compound of G.lucidum developed in this article could indeed improve sleep in mice induced by pentobarbital sodium,and can provide data to support the further researches in development of functional foods for improving sleep.
