ObjectsOsteoporosis is a bone disease with low bone mass and deterioration of bone structure,which has always been a threat to the public health of the elderly.At present,western medicine treatment has induced endometrial cancer,breast cancer,coronary heart disease and other side effects,however,studies have shown that traditional Chinese medicine has a definite therapeutic effect on osteoporosis,the Chinese medicine raw materials of health food stipulated by the ministry of health is safe,and the prescribed dose is less than the dosage of the drug,which can be taken for a long time,which has a better effect on the improvement of the symptoms of chronic diseases.The treatment of osteoporosis in traditional Chinese medicine is mainly warming and nourishing kidney yang,and withactivating blood circulation,however,in the present market,the traditional Chinese medicine health food of increasing bone density function lacksformula of tonifying kidney and activating blood circulation,therefore,it has a good market prospect to develop the traditional Chinese medicine health food of increasing bone density function withtonifying kidney and activating blood circulation.This research according to the ministry of health rules and a lot of literatures to screen the salt scurfpea fruit,salt eucommia,rhizoma drynariae,salvia miltiorrhizae,and calcined oyster.The combination of the drugs is warm,not dry,and the yin and yang are patched together,which can tonic kidney,strong bones,invigorate the circulation of blood and relieve pain.The purpose of this project is to develope a traditional Chinese medicine health food of increasing bone density function with reasonable and rigorous formula,reasonable and feasible process.Methods1.Research on the preparation processTo investigate the extraction factors such as the granularity of salt scurfpea fruit,the granularity of the calcined oyster,the time of extraction,the amount of water added,the number of extraction and the filtration method with weight rate of extract and psoralen and Isopsoralen,calcium content per dose as indicators,andthen investigate concentration,drying and preparation process.2,Research on the quality control methodsTo establishethe TLC identification method for salvia miltiorrhiza and rhizoma drynariae and the content determination method of psoralin and isopressin based on the Chinese pharmacopoeia of 2015,and then formulatethe draft quality standard.3.Research on the functional evaluationTo evaluate the functional evaluation of Guan according to increase bone density function test method of technical specification for health food inspection and evaluationissued by the ministry of health in 2003.The group was divided into sham operation group,model group,estrogen group,calcium carbonate group,low dose group,medium dose group and high dose group,each group of 10.The corresponding drugs were given 12 weeks to record weight.At the end of the experiment,the bone density was measured by micro-CT,and the content of bone calcium and bone phosphorus was determined by HPLC-ELSD method.Results1.Research on the preparation processThe best preparation:take the crushed salt scurfpea fruit and calcined oyster and other medicinal materials,extracting 3 times,for the first time with 10 times of water,extracting 60 min,the second time with 8 times of water,extracting 30 min,6 times for the third time in water for 30 min,filter by 20 mesh filter cloth and filtrate through 60℃ stress concentration to the density of 1.08 g/ml,then add soluble starch,dry by spray drying.Take dry paste,wet granule with 80%of ethanol,pressure,coating,make each piece 1.20g.2.Research on the quality control methodsThe method of TLC identification of salvia miltiorrhizaand and rhizoma drynariae and the determination method of psoralin and isopressin were established respectively.3.Research on the functional evaluationTheweight gain of high dose group and estrogen control group were significantly lower than the model control group(P<0.05),and the bone calcium,bone phosphorus content and bone mineral density of high dose group and estrogen control group were significantly higher than that of model control group(P<0.05).ConclusionThe formula of "Guan" is reasonable and rigorous.The preparation process is reasonable and feasible,and the quality is controllable.It has the effect of increasing bone density in the castrated rats.
