At present,with the development of our country’s economy,the dietary structure of urban residents has undergone great changes.The intake of high-energy foods whichfull of fat,sugar is increasing.All these high-calorie foods could induce the metabolic syndrome could induce glucose and lipid metabolism disorders,insulin resistance,type 2 diabetes and other metabolic syndromes which has become an important issue that threatens global health.In China,sesame is one of the traditional medicalfood.Sesame lignan is an active ingredient distributed in sesame,and has huge biological activities,such as stabilizing the human body’s redox balance and inhibiting cancer cell proliferation.Many resercher has studied the pure products of sesame lignans.The results has showed that: sesamin can improve the body metabolic disorder caused by high fat and high fructose diet;sesamin in sesame could not only eliminate active oxygen but also reduced free radical damage to mitochondria,and promote fatty acid oxidation,improve fat accumulation in the body.The pure products of sesame lignans has great effect of inhibit adipose,but few reports shows that whether the sesame extract has the ability to regulate lipid metabolism.Therefore,this study mainly investigate the regulation effect of sesame extract on obesity and fat-related lipid metabolism induced by high-fat diet and its possible mechanism.The main research contents and results are as follows:(1)Using ethanol-extract-method extract the sesame lignans and determine the lignans content.The ethanol-extract-method was: Using 85 % ethanol(assisted with ultrasound)extracte sesame 2 h,extraction temperature was 55 °C,ultrasonic power was 100 W,and solid-liquid ratio was 1:8.Under this method,the extraction rate of the extracted sesame extract was 0.11 %.Using high performance liquid chromatography(HPLC)determinethe contents of total lignans: sesamin was 3298.1 mg / 100 g,sesamol was 969.2 mg / 100 g,sasamol was 302.7 mg / 100 g.(2)High-fat diet (HFD) C57BL/6 mice was employed to study the inhibitory effects of sesame extract on obesity.The results was: sesame extract could not only reduce body weight gain,but also reduce serum triglyceride and cholesterol levels,what’s more could improve insulin resistance in mice.(3)After sacrificing mice,using H&E staining,PCR and other experimental methods to study the molecular-mechanism of sesame extract.a)In liver tissue,the sesame extract inhibited the expression levels of the lipogenic genes such as Pparg,Srebp1c,Fasn,and Acaca,and increased the lipolysismRNA levels of themitochondrial active genes such as Ppara,Cpt1a,Cpt2,Pgc1a,Sirt1,COXⅡ,and Tfam.Sesame extract can also improve liver cholesterol metabolism by down-regulating the express-ion of cholesterol synthesis genes HMGCR and ACAT2.b)In white adipose tissue(WAT),sesame extract also reduced mRNA levels of lipogenic genes Pparg,Srebp1 c,Fasn,and Acaca,and increased the mRNA levels of mitochondria-assoc-iated lipid metabolism genes Ppara,Pgc1a,Cpt1a,and Cpt2.promote.Improving Oxidation of fatty acids.c)In brown adipose tissue(BAT),sesame extract enhances the expression of mitochondrial biogenesis and heat-related genes Pgc1a,Ucp1,Fgf21,COXⅡ,and Tfam,there-by inhibiting browning of adipose tissue.In conclusion: The sesame extraction improved serum lipid levels,glucose tolerance,and abnormal tissue weight gain in mice.And inhibiting the expression of lipid metabolism related genes in the liver,white adipose tissue,and brown adipose tissue.Sesame extract has an inhibitory effect on mouse obesity.
