Bacteria drug resistance has been identified as one of the most important threats to human health in the 21st century by the World Health Organization.In recent years,drug resistance of foodborne bacteria has increasingly become the focus and research hotspot of international attention.In this study,a pathogenic bacterium,Vibrio parahaemolyticus,isolated from aquatic products,was used as the host bacteria for isolation and purification of bacteriophages,biological characteristics analysis,and then began the preliminary application study.First,Vibrio parahaemolyticus resistant strain Vp93 was isolated from Scophthalmus.It was used to screen bacteriophageΦVp3 andΦVp4 from sewage.After many times of depuration,we analyzed its biological characteristics.Finally,chose a bacteriophageΦVp4 as a sample to conduct research on purification function of Vibrio parahaemolyticus resistant strain in Scophthalmus maximus.Results show that bacteriophageΦVp4 can tolerate the high temperature of60°C,60°C lasts for 1 h and its effect price can reach 10~9PFU/m L;the range of p H tolerance was wide with higher activity between p H value 4~11,and the titer was up to 10~8 PFU/m L.The lyse spectra of the bacteriophage showed that the lyse rate of the bacteriophage could reach 0.41.Combined with the above results,the bacteriophage showed strong resistance to temperature and p H and a relatively wide cleavage spectrum,which could maintain a high titer under the growth conditions of its host bacteria Vibrio parahaemolyticus resistant strain Vp93,laying a theoretical foundation for the subsequent application research.The research conducted on purification function of bacteriophage for Vibrio parahaemolyticus resistant strain in Scophthalmus maximus has shown that breeding environment and the concentration of resistant strain of Vibrio parahaemolyticus obviously declined after adding the seawater infected by resistant strain of Vibrio parahaemolyticus to the bacteriophage,suggesting that the bacteriophage to drug-resistant bacterium purification of aquatic products is feasible;Found in the process of the experiment,according to the different concentration of resistant strain of Vibrio parahaemolyticus added,purification effect is slightly different.The main reason is forming an ecological balance between the bacteriophage and the host,causing bacterium cannot be completely removed.Therefore,multiple bacteriophage hybrid application of the“cocktail therapy”should be adopted in the purification of drug-resistant strain of Vibrio parahaemolyticus in Scophthalmus maximus when using bacteriophage or bacteriophage or other means of chemical or biological control act synergistically to effectively control Vibrio parahaemolyticus resistant strain and reduce its harm for food safety and human health.
