Auxis rochei,a kind of low-value marine fish with the smallest body in the tuna family,however,it has rich nutritional value and high meat quality.It is widely distributed in China and can be fished almost all the year round,with extremely high economic value.At present,the yield of skipjack is increasing year by year.In this paper,we used bacillus subtilis protease as raw material to obtain Auxis rochei hydrolysates from Auxis rochei rutabilis by vacuum concentration and spray drying.Through animal experiments,we investigated the in vivo anti-fatigue activity and uric acid lowering activity of bonito hydrolysates from Auxis rochei rutabilis.To establish a method for detecting the inhibition rate of angiotensin converting enzyme by high performance liquid chromatography(HPLC),and to explore the in vitro blood-pressure-lowering activity of Auxis rochei hydrolysates,which will provide a new reference for the comprehensive development and utilization of A uxis rochei.1.The results of the anti-fatigue experiment on mice showed that:the bubonito hydrolysates made by gavage had no significant effect on the normal weight increase of male mice with SPF KM,and the swimming times of the mice with exhaustion were all prolonged,among which the low and medium dose groups increased by 89.24%and 92.24%respectively,and the difference was extremely significant(p<0.01).Compared with the control group,the content of urea nitrogen in the low,medium and high dose groups decreased by 20.02%,17.54%and 18.52%,respectively.Compared with the blank control group,the serum lactic acid content in the low,medium and high dose groups of Auxis rochei hydrolysates decreased by 29.30%,33.37%and 46.71%,respectively,and the difference between the dose groups and the blank control group was extremely significant(p<0.01).The contents of glycogen in the liver of mice in the low,medium and high dose groups were significantly increased(p<0.05)(increased by 45.88%,43.81%and 47.99%,respectively).The results showed that the bonito hydrolysates showed good anti-fatigue activity in vivo.2.Uric acid lowering test results on mice showed that:the Auxis rochei hydrolysates produced by intragtragically gavage had no significant effect on the normal weight increase of male mice with SPF ICR;Compared with the model control group,the effects of low,medium and high dose groups on serum uric acid(UA),blood urea nitrogen(BUN),creatinine(CRE),liver XOD activity and adenosine deaminase(ADA)activity were differect.Serum uric acid UA values of the mice were significantly lower than those of the model group(decreased by 55.27%,33.57%and 24.86%,respectively).In the liver of mice,XOD values were reduced by 23.67%,30.72%,18.86%,and ADA values were decreased by 49.09%,33.88%,and 34.15%,respectively,showing extremely significant differences(p<0.01).Low and high dose groupsshowed no significant difference in serum BUN level(p>0.05),and no significant difference in CRE value(p>0.05).The results showed that the Auxis rochei hydrolysates had the activity of decreasing uric acid in vivo.3.The inhibition rate of ACE was determined by HPLC in vitro.High performance liquid chromatography(HPLC)was developed to detect the ACE inhibition rate.The influence of the mobile phase and the order of sample addition in the detection system was discussed.The results showed that the ACE inhibition rate was determined by HPLC,and the IC50 was 27.08 μg/mL,and the IC50 of captopril was 1.84×10-3 μg/mL.The results showed that Auxis rochei ACE inhibitory peptides had certain effect in vitro activities of lowering blood pressure.
