The Mongolian medicine Corydalis is the tuber of Corydalis yanhusuo W.T.Wang,a plant in the poppy family,which has analgesic,sedative and hypnotic effects.Modern pharmacological experiments have proved that alkaloids of this medicine are its main active ingredients.In this paper,Mongolian medicine Corydalis was taken as the research object,its compounds were extracted and their structures were identified,alkaloid compounds were prepared by HSCCC and compounds with anti-inflammatory effects were screened by molecular docking technology,providing experimental basis for in-depth study of Mongolian medicine Corydalis,the contents are as follows:(1)Mongolian medicine Corydalis was extracted with ethanol,and then petroleum ether,ethyl acetate and n-butanol were successively used to extract the ethanol extract.Using silica gel column chromatography,Thin layer chromatography and liquid chromatography,a total of 19 compounds were isolated from the ethyl acetate extract of Mongolian medicine Corydalis,and 13 compounds were identified,which were as follows: d-corydaline,tetrahydropalmatine,dehydrocorydaline,palmatine,tetrahydroberberine,protopine,tetrahydrocoptisine,tetrahydrocorysamine,glaucine,dehydroglaucine,isocorybulbine,tetraydrocolumbamine and β-sitosterol.(2)The tetrahydrocorydinine and dehydrocorydinine in Mongolian medicine Corydalis were purified by HSCCC.The total alkaloids of Mongolian medicine Corydalis were prepared by ionic resin,and an ULTRA-performance liquid chromatography(UHPLC)method was established.The conditions were as follows:mobile phase 70% methanol aqueous solution,flow rate 0.3 m L/min,column temperature 40℃.The solvent system of high speed countercurrent chromatography by liquid chromatography is petroleum ether-ethyl acetate-methanol – water(16:31:19:21,v/v/v/v).In this solvent system,the sample volume was 0.5 g,the flow rate was 2.0 m L/min,the detection wavelength was 280 nm,and the speed was900 RPM.The purity of tetrahydrocorydinine and dehydrocorydinine were 95.10%and 91.20%.(3)Molecular docking technology was adopted to explore the effective components of the anti-inflammatory effect of Mongolian medicine Corydalis.Among them,the proto-berberine alkaloids tetrahydrotilberine,dehydrocorydaline,apomorphine alkaloids glaucine and steroidal compounds β-sitosterol showed high affinity with anti-inflammatory proteins COX-2,1S1 R and 1DVX,which showed potential anti-inflammatory activity.Therefore,the anti-inflammatory effect of Mongolian medicine Corydalis presents a mechanism of multi-component and multi-target action.
