B vitamins are a class of organic compounds that are indispensable to the normal activities of the human body.Except for vitamin B12,other B vitamins cannot be directly synthesized by the human body and must be obtained from food.When the human body lacks or consumes too much B vitamins,it will produce angular cheilitis,dermatitis and other diseases.The content of B vitamins in food is low and unstable,and there are matrix interferences such as high sugar and high starch,which affect the accuracy of test results.Therefore,it is necessary to establish a rapid,accurate,and low-matrix interference method for the detection of B vitamins.In this work,a rapid method for the detection of B vitamins was developed using two-dimensional carbon fiber liquid chromatography(2DμCFs)tandem electrospray mass spectrometry(ESI-MS).Using the carbon fiber material with good stability and large difference in surface properties as the stationary phase,the complex samples were divided into three polar components:strong,medium and weak,which were then entered into electrospray mass spectrometry.Its characteristics are that the matrix interference between different components is reduced,and the ionization suppression effect is also reduced,and the sensitivity of mass spectrometry analysis is improved.By combining 2DμCFs with ESI-MS,the comprehensive analysis and quantitative detection of B vitamins in samples can be realized,in order to provide technical support for the quality evaluation of B vitamins in food.Based on the above,the work carried out is as follows:1.The detection method of 8 kinds of B vitamins(vitamin B1,vitamin B2,vitamin B3,niacinamide,vitamin B5,vitamin B6,vitamin B9,vitamin B12)was established by2DμCFs-Qq Q-MS/MS,and 8 kinds of B vitamins were realized within 5 minutes.Isolation and detection of targets.The method has a good linear range,R2>0.9976,with good sensitivity(LOD=1.7 ng m L-1;LOQ=5.5 ng m L-1)and repeatability(RSD<15%).2.This method is applied to the detection of B vitamins in B vitamin tablets and cauliflower.The sample was extracted with 0.01 mol L-1 HCl as solvent and ultrasonically.After centrifugal filtration,the extract was directly sent to 2DμCFs-Qq Q-MS/MS to separate B vitamins of different polarities,and at the same time remove weakly polar pigments,lipids,etc.Interfering substances,qualitative and quantitative detection of the target.The results showed that the content of B vitamins in B vitamin tablets was within a reasonable range compared with the standard value,and the content of B vitamins in cauliflower was 0.076-0.299μg m L-1.The method can be applied to the screening and rapid quantitative detection of B vitamins in complex matrix samples.3.Taking advantage of the fast and easy operation of this method,2DμCFs combined with QTOF-HRMS was used to comprehensively analyze the chemical composition of B vitamin tablets.Provide technical support for quality assurance of B vitamins in screening foods.
