Objective:To determine the best wall-breaking process of rape bee pollen using single-factor experiments,and to investigate the physicochemical properties of the active peptides of rape bee pollen,the effects on hypoxia-damaged cells,and the effects on the motility of mice,in order to provide a theoretical basis for the optimization of the wall-breaking process of rape bee pollen and the research as a functional food.Methods:One-way ANOVA and orthogonal test were used to determine the optimal conditions for breaking the wall of rape bee pollen.The antioxidant capacity of the peptides was measured in vitro by DPPH and ABTS radical scavenging assay;the effect of the peptides on the cells damaged by hypoxia was investigated by establishing a hypoxic injury model using Co Cl2;finally,different doses of the peptides were given to mice by gavage to study the effect on their locomotor ability.Results:1.Results of the best wall-breaking process of rape bee pollen:after 4days of lactic acid bacteria liquid fermentation,15 min of ultrasound at 480 W output was the best condition for wall-breaking,and the protein extraction rate reached 10.7%under this condition.2.Results of physical and chemical property determination of rape bee pollen active peptide:after enzymatic digestion,rape bee pollen protein had been fully hydrolyzed into small molecule active peptide,and the solubility of fermented group was significantly higher than that of unfermented group.The solubility of the fermented group was significantly higher than that of the unfermented group.3.Antioxidant in vitro free radical scavenging experiment:the free radical scavenging ability of the active peptide was positively correlated with the content of the active peptide.4.Effect of active peptide on cells damaged by hypoxia:the cell survival rate of the fermented group was higher than that of the unfermented group;when the action time of Co Cl2 was less than 12h,the cell survival rate was positively correlated with the concentration of the active peptide;the longer the action time of Co Cl2,the longer the effect on cell hypoxia.The longer the action time,the weaker the protective effect on cellular hypoxic damage.5.The effect of different concentrations of active peptide of rape bee pollen on the exercise ability of mice:there was no significant effect of active peptide of rape bee pollen on the body weight of mice,after long time exercise training,the high concentration of active peptide of rape bee pollen could reduce the number of mice falling off the running platform and improve the exercise ability of mice.Conclusion:1.The best conditions for breaking the wall of rape bee pollen were:4 days of liquid fermentation by lactic acid bacteria,480 W of ultrasonic power and 15 min of ultrasonic time.2.The active peptides of rape bee pollen after microbial fermentation had smaller molecular weight and better water solubility,which were easily absorbed by the organism.3.It was concluded that the active peptide of rape bee pollen can effectively relieve exercise fatigue and enhance exercise capacity,which can be used for the development of functional food.
