Acne is a chronic inflammation that occurs in the sebaceous glands and hair follicles.For the treatment of acne,compared with antibiotics,people are paying more and more attention to the acne-removing effects of natural plant-derived compounds.Kapok is mainly distributed in southern my country.In addition to its ornamental value,kapok is also rich in chemical components,including flavonoids,organic acids,steroids,triterpenes and other chemical components,of which flavonoids are the most abundant.In this paper,kapok was selected as raw material,the flavonoids in kapok were extracted by ultrasonic-assisted alcohol extraction technology,and the extracted kapok flavonoids were purified by macroporous resin.The oxidative activity and whitening activity were studied,and they were applied in cosmetics to study their acne-removing effects.The main research contents and conclusions are as follows:(1)Ultrasound-assisted alcohol extraction technology was used to extract kapok flavonoids.On the basis of single factor experiment,the optimal extraction process of kapok flavonoids was optimized by orthogonal experiment.60,the extraction time was 40 min,the volume fraction of ethanol was 45%,and the extraction temperature was 70°C.Under this condition,the yield of kapok flavone was 10.07±0.29%.(2)Macroporous resin was used to purify kapok flavonoids.By comparing the adsorption and desorption rates of 8 macroporous resins on kapok flavone,it was found that D101 had the best effect,and the desorption effect was when the volume concentration of desorption liquid ethanol was 60%.The best;the optimal dynamic adsorption-analytical conditions are:the sample volume is 2 BV,and the elution volume of the analytical solution is0.625 BV.After purification,the purity of kapok flavone increased from 32.5%to 98.2%.With the help of HPLC-ESI-MS combined analysis technology,40 components were separated and identified,of which the most abundant components were isomgiferin,quinic acid and quercetin-3-glucuronide.The relative contents of these three components were the same.The sum is 64.56%.(3)The antioxidative activity of kapok flavonoids was studied,and the free radical scavenging experiments of ABTS·+and DPPH showed that kapok flavone had strong antioxidant capacity,and its effect was only slightly lower than that of the positive drug V_C.The purified kapok flavone has excellent whitening effect,and has a very significant inhibitory effect on tyrosinase that leads to the production of melanin.The inhibition rate of kapok flavone on tyrosinase is higher than that ofα-arbutin.Through the zebrafish embryo melanin inhibition experiment,it can be seen that when the purified kapok flavone concentration is 1 g/L,the inhibition rate of zebrafish embryo melanin is 12%,and it has significant whitening activity.The antibacterial effect of kapok flavonoids was explored through the inhibition zone experiment.When the concentration of kapok flavonoids was 12.5g/L,the diameter of the inhibition zone against Staphylococcus epidermidis was 20.33±0.47 mm,which showed high sensitivity and was highly sensitive to P.acnes.The diameter of the inhibition zone was 11.67±0.47 mm,showing moderate sensitivity.Mouse macrophage Raw264.7 was used to establish an inflammation model of TNF-αand IL-6 to determine the anti-inflammatory effect of purified kapok flavonoids.,has an inhibitory effect on the production of the pro-inflammatory factor TNF-α,and the inhibitory effect is dose-dependent.When the concentration of kapok flavone was 0.25~1.0 g/L,it had inhibitory effect on the production of pro-inflammatory factor IL-6,and the inhibitory effect showed a dose-dependent relationship,and when the mass concentration of kapok flavone solution was1.0 g/L,the inhibitory effect on IL-6 was equivalent to that of the positive control Oxumin(1%).(4)According to the results of the in vitro anti-acne efficacy experiment,the kapok flavonoid extract was added to the basic cream for human testing.After the acne subjects used the kapok flavone cream for 28 days,the skin brightness,oil secretion and TEWL were all improved.The inflammatory status of the face showed a downward trend.Through 16Sr DNA sequencing technology,it was measured that after using the kapok flavonoid cream for 28days,the content of P.acnes on the face of the subjects with acne decreased,the flora richness increased,and the skin flora increased.The imbalance is improved.
