| Hazelnut oil contains more than 90%of unsaturated fatty acids,and contains vitamin E,sterol,squalene and antioxidant phenol.It is a kind of functional oil with great edible and research value.In this study,hazelnut oil was used as raw material to determine its main components,physicochemical properties and antioxidant activity in vitro and in vivo.The main results were as follows:1)The fatty acid composition and physicochemical properties of the oils of Co-rylus heterophylla,Corylus mandshurica and hybrid hazelnuts were determined,inclu ding acid value,iodine value,saponification value and peroxide value.The results s-howed that the contents of unsaturated fatty acids and oleic acids in the three haze-lnut oils were all as high as 95%and 75%respectively.Physical and chemical pro-perties.Each index accords with edible oil standard.acid value:C.mandshurica<C.heterophylla<hybrid hazelnuts;iodine value:C.heterophylla<hybridhazelnuts<C.mandshurica;peroxide value:C.mandshurica<hybridhazelnuts<C.heterophylla;s-aponification value:hybrid hazelnuts<C.heterophylla<C.mandshurica.2)The antioxidant activity of the oils of C.heterophylla,C.mandshurica and hybrid hazelnuts.in vitro was studied.The free radical scavenging capacity,hydroxyl radical scavenging capacity and iron ion reducing capacity of the three oils of hazelnuts were compared.Within the range of 5~25 mg/m L,the higher the oil concentration was,the stronger the antioxidant capacity was,showing a dose-effect relationship.When the concentration reached 25 mg/ml,the DPPH free radical scavenging rate of C.mandshurica oil was 45.21%,DPPH free radical scavenging rate of C.heterophylla oil was 23.7%,DPPH free radical scavenging rate of hybrid hazelnuts oil was 20.2%,Hydroxy radical scavenging rate of C.mandshurica oil was 88.22%,Hydroxy radical scavenging rate of Corylus heterophylla oil was 78.86%,Hydroxy radical scavenging rate of hybrid hazelnuts oil was 52.47%,and the total reduction capacity of C.mandshurica oil is higher than the other two oils.3)The antioxidative damage model of L929 cells induced by H2O2 was established,and the protective effect of three kinds of hazel oil on oxidative damage cells was studied.The results showed that the hazel oil of 25~400μg/m L did not promote or inhibit the growth of normal L929 cells.The oxidative damage model of L929 cells induced by H2O2with the concentration of 200μmol/L was established,and the survival rate of oil oxidation-damaged cells in the Corylus mandshurica group increased from 50.54±2.72%to 79.65±0.41%,which was statistically significant(P<0.01).The results showed that after the cells were treated with a concentration of 400μg/m Lof Corylus mandshurica,the fluorescence product 2′,7′-dichlorofluorescein(DCF)decreased significantly compared with the oxidative damage model group,indicating that the active oxygen(ROS)content was also significantly lower,and the effect of the Corylus mandshurica and hybrid hazelnuts oils on the ROS content of oxidative damage L929 cells was not significant.Therefore,the strongest anti-oxidative ability of Corylus heterophylla oil was screened out as hazel oil.4)To sum up,Corylus mandshuric oil was selected for further antioxidant activity in vivo,and the aging model of mice induced by D-galactose(D-gal)was established to detect the effect of low,medium and high dose of Corylus avellana oil on aging model mice.Effects of weight,organ index,and biochemical markers of serum,liver,heart,kidney and brain,such as malondialdehyde(MDA)content,superoxide dismutase(SOD)activity,glutathione peroxidase(GPX)activity.The results showed that hazelnut oil had a protective effect on aging and oxidative damage induced by D-gal.Hazelnut oil could maintain the body weight of aging mice(P<0.05),improve the atrophy of liver and spleen induced by D-gal,and increase the activity of T-AOC,SOD and GPX in serum and viscera of aging mice(P<0.05),and decrease the content of MDA in serum and viscera of aging mice(P<0.05). |
