Fibularhizoctonia sp.is a symbiotic fungus found in scattered termite nests.This thesis takes termite balls as the research object,with adenosine as the index of termite balls shake flask fermentation liquid volume was optimized and conducted a 7-day cycle experiment to select the best fermentation time.On this basis,a fermenter fermentation experiment was conducted on termite balls using a 5 L fermenter for the first time,and the resulting mycelial powder was investigated for animal hypolipidemic function,which may provide a basis for pilot scale-up of termite balls,and a foundation for the development and utilization of termite balls mycelial powder related products.The main results are as follows.(1)The experiment of optimizing the fermentation conditions of termite ball in medicine bottles was carried out.Taking adenosine as the index,the amount of shake flask solution of termite cocci was optimized.On this basis,a 7-day cycle experiment was carried out.The dry weight of termite ball mycelium,adenosine content,p H value and residual sugar concentration of fermentation solution within 7 days were detected to confirm the best time for termite ball to produce adenosine,and the extraction time of termite ball shake flask fermentation seed solution and adenosine.The results showed that the amount of fermentation liquid was 15%,and the content of adenosine reached the maximum on the third day of fermentation,up to 4.45 mg/g;Using the fermentation broth as the fermentation seed liquid of termite ball was more conducive to the synthesis of adenosine by termite balls,and the content of adenosine was 5.17 mg/g;The best extraction time of termite balls by ultrasonic water extraction is 3h.(2)Termite balls 5 L fermenter experients is a scale-up experiment using a 5 L fermenter for termite ball on the basis of shake flask fermentation.A single factor experiment was used to initially explore the stirring speed,ventilation volume and inoculation volume.The experimental results show that the 5 L fermentor of termite balls can be fermented by using a stirring speed of 100 r/m,an aeration volume of 3 l/m,and a bottle of seed liquid.(3)According to the national standard,the detection results of heavy metal ions and aflatoxin B1in termite mycelium powder showed that the content of lead in mycelium powder was 0.11mg/kg,arsenic content was less than 0.085 mg/kg,mercury content was 0.047 mg/kg,cadmium content was 0.017 mg/kg,aflatoxin B1was less than 0.03μg/kg.In line with the national edible fungus safety standards.(4)Using SD rats as experiental animls to construct a high-fat model of SD rats using a high-fat diet,the suspension prepared from termite balls was set at 1.00 g/kg.bw,0.50 g/kg.bw,0.25 g/kg.bw,as high,medium and low three dose groups,fundus venous blood collection after 30 days of gavage was performed to measure TC,TG,LDL-C,and HDL-C in serum.The results of this experient are as follows,termite balls hyphae powder decreased serum triglyceride TG in the model rats,but the difference did not reach significance when compared with the control group.In conclusion,in this study,the production of adenosine of termite balls was increased to 5.17mg/g by shaking flask fermentation,and the maximum production of adenosine was 5.79mg/g after the 5 L fermentation tank experiment of termite balls.In addition,the heavy metal elements and aflatoxin in mycelium powder met the hygienic requirements of edible fungi,laying a foundation for the industrial production of termite balls.
